氯胺酮对小鼠短暂性局灶性脑缺血再灌注损伤的影响

摘    要:目的:观察不同非麻醉剂量氯胺酮对小鼠短暂性局灶性脑缺血性再灌注损伤的影响.方法:通过栓塞大脑中动脉制备小鼠短暂性局灶性脑缺血再灌注模型,缺血2 h再灌注,以神经行为评分达Bederson's 3级视为模型成功标准,再灌注24h处死取全脑进行TTC染色,测量梗死面积,HE染色观察海马病理变化.结果:25mg/kg氯胺酮能显著减少脑缺血再灌注损伤引起的梗死面积(P<0.05)和改善海马神经细胞坏死,而10mg/kg和15 mg/kg氯胺酮无统计学意义,但呈明显剂量依赖性关系.结论:非麻醉剂量氯胺酮能改善小鼠短暂性局灶性脑缺血再灌注损伤并呈明显剂量依赖性关系.

Abstract:Objective:To study effects of different sub-anaesthetic doses of ketamine on reperfusion damage after transient middle cerebral artery occlusion in mice.Methods:Mice subjected to transient middle cerebral artery occlusion, neurological evalution was conducted at 2 hr after occlusion, according to the Bedersons standard, 3 rank was viewed as standard of successful model, All mice sarcrificed at 24 hr after reperfusion for the determination of infarction volume (%) and examination of parhological changes.Results:25mg/kg ketamine attenuated infarction volume (%)significantly (P<0.05)and improved pathological changes to some extent , all doses showed dose-dependent action.Conclusion:Ketamine is able to improve the damage after transient middle cerebral ischemia-reperfusion, and shows dose-dependent action.
