摘    要:目的建立同时测定覆盆子中3种黄酮成分含量的超高效液相色谱法(ultra performance liquidchromatography,UPLC)。方法采用BEH C18(2.1mm×100mm,1.7μm)色谱柱;以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱;流速0.12ml/min,检测波长365nm,柱温20℃。结果椴树苷、槲皮素、芦丁分别在1.31~13.08、1.92~19.20、49.20~492.00ng范围内与峰面积呈现良好的线性关系。平均加样回收率分别为99.7%、99.7%、100.2%,相对标准偏差分别为1.03%、0.74%、1.42%。不同产地和采收期覆盆子中3种黄酮成分含量有显著差异。结论该方法简单、可行,可作为覆盆子药材质量控制的参考依据。

Simultaneous Determination of Three Flavonoids in Fructus Rubi by Ultra Performance Liquid Chromatography

Abstract:Objective To establish a method for determining the contents of three flavonoids in Fructus Rubi by ultra performance liquid chromatography (UPLC). Methods UPLC determination was carried out on a BEH C18 chromatographic column (2.1mm×100mm,1.7μm) with a mobile phase of acetoni trile-0.1%0 phosphoric acid (for gradient elution) at a flow rate of 0.12 ml/min, a detection wavelength of 365 nm, and a column temperature of 20℃ to detect three flavonoids (linden glycosides, quereetin, and ru toside). Results The contents of linden glycosides, quercetin, and rutoside had a good linear relationship with the peak areas within the ranges of 1.31 13.08 ng, 1.92-19.20 ng, and 49.20-492.00 ng, respective- ly. Their average recovery rates were 99.7%, 99.7%, and 100.2%, respectively, and the relative stand- ard deviations were 1.03%, 0.74%, and 1.42%, respectively. The contents of three flavonoids in Fructus Rubi produced in different places and harvested at different times were significantly different. Conclusion The UPLC determination method can be used for the quality control of Fructus Rubi because of its simplicity and feasibility.
