人脐带间充质干细胞分离培养方法的优化

摘    要:目的:优化人脐带间充质干细胞 ( umbilical cord mesenchymal stem cells, UC - MSCs)分离培养的方法。方法:无菌条件下剔除剖腹产胎儿脐带动静脉,用牙剪子剪为约lmm。但彼此相连的组织块,得到贴壁细胞,采用半量换液进行原代及传代培养。用流式细胞仪检测其免疫表型。结果:成功从人脐带中分离纯化得到UC—MSCs,呈条形和成纤维细胞样的两种形态。UC—MSCs高度表达CD105、CD73.CD44、CDg0,极少数表达CIM5、CD34-CD14和HLA—DR。结论:建立出一种快捷经济的UC—MSCs体外分离培养方法。

OPTIMIZATION OF ISOLASION AND CULTURE METHODS OF HUMAN UMBILICAL CORD- DERIVED MESENCHYMAL STEM CELLS

Abstract:Objective:To optimize the isolation and expansion system of umbilieaI cord -derived mesenchymal stem cells (UC - MSCs). Methods: Under sterile conditions, umbilical artery and vein were excluded from cesarean section fetal umbilical cord. With the toothed scissors, the umbilical cord was cut into i mm3 tissue blocks but are connected to each other. After getting adherent cells, they were used to primary and passage culture by changing half of the medium. The immunophenotype of UC - MSCs was detected by flow cytometry. Results: UC - MSCs was successfully/isolated from human umbilical cord, and there are two types of strip sample and fibroblast - like. Using flow cytometry, all analyzed UC - MSCs were labeled positively for CD105, CD73, CD44 and CD90, but nega- tively for CD45, CD3d, CD14, and HLA- DR. Coneluslon: A simple and convenient method to isolate and culture UC- MSCs in vitro was successfully established.
