宁夏滩羊KAP1.1基因多态性及其与二毛裘皮重要性状关系的研究

摘    要:目的:研究宁夏滩羊KAP1.1基因的遗传多态性及其多态位点与滩羊二毛毛股弯曲数、纤维直径及毛股自然长度的关系,寻找可用于辅助选择的分子标记,为下一步分子育种提供理论依据。方法:采用PCR—SSCP技术研究了261只滩羔羊KAPl.1基因的多态性,利用DPS7.05软件分析了2个多态位点不同基因型裘皮性状的最小二乘均值(LSM),并进行了多重比较。结果:发现KAPl.1基因有两个突变位点sl和s2,其中s1位点有3个基因型,S2位点有5个基因型。在二毛裘皮毛股弯曲数性状上,S1-AA的LSM高于S1-AB、S1-BB,但三种基因型阃无显著差异(P〉0.05),S2-AB的LSM高于S2-AA、S2-BB、S2-AC、S2-DD,但五种基因型间无显著差异(P〉0.05);在毛股自然长度性状上,S1-AB的LSM显著高于S1-BB(P〈0.05),S2-A基因型的LSM板显著高于S2-BB(〈.);在毛纤维细度性状上,S1-AA的低于S1-AB、.CP001LSNABs1一BB,但三种基因型阃无显著差异(P〉0.05)。结论:本研究发现了宁夏滩羊KAP1.1基因的两个多肽位点S1和S2,其中S1位点有3个基因型,而s2位点有5个基因型,处于高度多态(PIC=0.7078)和高度杂合(h=0.7078),该位点上遗传多样性很丰富,在品系培育上选择潜力很大;S1位点的AB基因型和s2位点的AC基因型都与滩羊二毛皮的毛股自然长度有明显关系,可作为分子标记用于二毛裘皮较长毛股自然长度潍羊品系定向选育。

THE STUDY ON KAP1.1 GENE POLYMORPHISM AND RELATIONSHIP BETWEEN THE GENE POYYMORPHISM AND LAMB INPORTANT FUR TRAITS OF NINGXIA TAN SHEEP

Abstract:Objective:We study the polymorphism of Ningxia Tan KAP1.1 gene and the relationship between the gene polymor- phis'm and fur important traits of lamb, including wool fiber diameter, the number of bending and the natural length of wool stocks in Order to discover molecular - assisted selection mark for molecular breeding in the following work. Methods: PCR - SSCP technique was used to analyse KAP1. 1 gene polymorphism of 261 lambs. Then, software DPS7.05 was used to analyse the least squares mean (LSM) of fur traits in two different genotypes~ and multiple comparisons of them ere carried out. Results:We found that KAP1. l gene has S1 and S2 two mutations, in which S1 locus has three genotypes, and S2 locus has five genotypes. In number of bending of wool stocks traits, the LSM of S1 - AA is higher than S1 - AB and S1 - BB, but there was no 'significant difference among the three genc~ types ( P 〉 0.05 ) ; The LSM of S2 - AB is higher than S2 - AA , S2 - BB, S2 - AC and S2 - DD, but there was no significant differ- ence among the five kinds of genotypes ( P 〉 0.05 ). In natural length of wool stocks traits, the LSM of S1 - AB was significantly high- er than S1 - BB ( P 〈 0.05 ), and the ISM of S2 - AC was significantly higher than S2 - BB ( P 〈 0.01 ), In the wool fiber fineness traits, the LSM of S1 - AA is lower than S1 - AB and S1 - BB, but no significant difference was among three genotypes ( P 〉 0.05 ). Conclusion: KAP1.1 gene in Ningxia Tan sheep has two peptides S1 and S2, in which S1 locus has three genotypes, and S2 site has five genotypes, while S2 site is highly polymorphic ( PIC = 0.7078 ) and highly heterozygous ( h = 0.7078), in which genetic diversity is very rich and conducive to breed new strain; AB genotype with S1 site and AC genotype with S2 site both are clearly related to the natural length of wool stocks in Tan sheep, and the two sites can be used as molecular markers to breed Tan sheep strain with higher natural length of wool stocks.
