哮喘患儿外周血CD4+CD25+调节性T细胞的变化及其与哮喘病情的关系——附80例检测报告

摘    要:目的:探讨哮喘患儿外周血CD4+CD25+调节性T细胞(regulatoryTcell,Treg)的变化及其与哮喘病情的关系。方法:80例哮喘患儿(哮喘组)按,临床表现分为急性发作期组(60例)和缓解期组(20例),其中急性发作期组按病情再分为轻度组(34例)和中度组(26例),另选择50名健康儿童为正常对照组。应用流式细胞仪检测上述各组外周血CD4+CD25+Treg细胞水平(CD4+CD25+Treg占CD4+T细胞的百分比),采用整群和分层研究的方法,进行统计学分析。结果:哮喘组CD4+CD25+Treg水平明显低于正常对照组(P〈0.01)。急性发作期组CD4+CD25+Treg水平明显低于缓解期组和正常对照组(均为P〈0.05)。轻度组CD4+CD25+Treg水平则明显高于中度组(P〈0.01)。经Spearman等级相关分析,哮喘患儿外周血CD4+CD25+Treg水平与哮喘的病情严重程度呈负相关(r=-0.361,P〈0.01)。结论:与健康儿童比较,哮喘患儿外周血CD4+CD25+Treg水平明显降低,而在哮喘发作期降低更甚。随着哮喘患儿病情的加重,其外周血CD4+CD25+Treg水平逐渐降低。

The changes of CD4+CD25+ regulatory T cells in peripheral blood and their significance in the disease severity of asthmatic children

Abstract:Objective: To investigate the changes of CD4 + CD25 + regulatory T cells (Treg) in peripheral blood and their significance in the severity of asthmatic children. Methods: Eighty asthmatic children (asthma group) were divided into acute attack stage group (60 cases) and remission stage group (20 cases) according to their clinical features, and the acute attack children were divided into the mild asthma group (34 cases) and the moderate asthma group (26 cases) according to their severity of asthma. Meanwhile, fifty health children were en- rolled as control group. The levels of CIM + CD25 + Treg in peripheral blood of all children were tested by flow cy- tometer(FCM). Cluster study and stratified study were statistically analyzed. Results: The levels of CD4 + CD25 + Treg of asthmatic children were lower than those in the control group( P 〈0. 01 ). The CD4 + CD25 + Treg levels in acute attack group were lower than those in remission group or in the control group (P 〈 0. 05, in each). The CD4 + CD25 + Treg levels in the mild group were higher than in the moderate group( P 〈 0.01 ). There was remark- ably negative correlation between CD4 + CD25 + Treg levels and the severity of asthmatic children by Speraman cor- relation coefficients analysis ( r = -0. 361, P 〈0. 01 ). Conclusion: The levels of CD4 + CD25 + Treg are remarka- ble decrease in asthmatic children and drastic decrease in attack asthma compared with the control group. The more severity of asthma, the more decrease of levels of CD4 + CD25 + Treg in peripheral blood in asthmatic children.
