参麦注射液对大鼠心肌缺血再灌注损伤的保护效应及其与热休克蛋白的关系

摘    要:目的 :研究参麦注射液对大鼠心肌缺血再灌注损伤的保护效应以及与热休克蛋白 (HSP70 )的关系。方法 :用在体左冠状动脉前降支穿线结扎法制备心肌缺血再灌注模型。 60只SD大鼠分为三组 ,假手术组n =10只 ,缺血再灌注组n =2 5只 ,参麦注射液干预组n =2 5只。缺血 3 0min ,再灌注 12 0min。观察心肌梗死范围和心肌细胞超微结构变化 ,采用S P免疫组化法 ,检测HSP70 的表达。结果 :参麦注射液干预组与缺血再灌注组相比 ,缺血面积 ( % ) ( 3 8.3 6± 2 .62vs 42 .3 2±2 .2 8,P <0 .0 1) ;梗死面积 ( % ) ( 59.3 6± 2 .44vs 65.63± 1.68,P <0 .0 1)。HSP70 表达 ( 0 .13 8± 0 .0 2 2vs 0 .12 2± 0 .0 18,P <0 .0 1)。电镜下 ,缺血再灌注组 ,肌纤维挛缩 ,核固缩 ,线粒体嵴疏松 ,空泡形成。参麦注射液干预后 ,肌节清晰 ,线粒体嵴较密集 ,无明显空泡形成。结论 :参麦注射液能保护心肌超微结构 ,缩小缺血、梗死范围 ,其机制可能与HSP70 的表达增加有关

Abstract:To sutdy protective effects of SheMai injection on myocardium ischemia reperfusion(I/R) injury in rats and expression of HSP 70 .Method: Myocardial ischemia reperfusion injury model was induce d by left descending anterior coronary artery(LAD) ligation in 30 min,followed b y 120 min of reperfusion in vitro.60 SD rats were divided into three groups:Sham group;I/R group;ShenMai group.The size of myocardial infarct and the changes of ultrastruc ture of myocardium had been investigated.The expression of HSP 70 was exam ined by s p immunohistochemical.Results: The xize of myocardial ischemic and infarct and the exp ression of HSP 70 in ShenMai group were compared with that in I/R group:38. 36±2.62 vs 42.43±2.28,59.36±2.44 vs 65.63±1.68,0.138±0.022 vs 0.122±0.018, P<0.01 ,respectively.Electron microscope:contracted myofibrilla,rarefied mito chondrial spine and large vacuoles in mitochondrial were detected in I/R group,b ut clear sarcomere,dense mitochondrial spines and no obviously vacuoles in mitoc hondrial was observed in ShenMai group.Conclusion: The ultrastructure of myocardium and the size of isc hemic and infarct could be protected and reduced by ShenMai injection in the cou rse of ischemic reperfusion injury.The mechanism of cardioprotection may be ass ociated with the expression of HSP 70 in myocardium.
