异硫氰酸苯乙酯对人乳腺癌MCF-7细胞凋亡作用及机制研究

摘    要:目的探讨异硫氰酸苯乙酯诱导人乳腺癌MCF-7细胞凋亡的作用及其可能的机制。方法以不同浓度异硫氰酸苯乙酯处理人乳腺癌MCF-7细胞,应用MTS法观察异硫氰酸苯乙酯对MCF-7细胞的抑制率,流式细胞术检测异硫氰酸苯乙酯对细胞凋亡、细胞周期以及活性氧表达的影响。结果将不同浓度的异硫氰酸苯乙酯作用于MCF-7细胞72h后,抑制了MCF-7细胞的增殖,其IC50为15.2μmol.L-1;以10和20μmol.L-1的异硫氰酸苯乙酯作用MCF-7细胞24h后,细胞出现凋亡现象,细胞周期阻滞在G2/M期,并造成细胞氧化损伤。结论异硫氰酸苯乙酯能够促进人乳腺癌MCF-7细胞凋亡,影响细胞周期,其作用机制可能与异硫氰酸苯乙酯可造成肿瘤细胞氧化损伤有关。

Abstract:Objective To investigate the effect and mechanism of phenethyl isothiocyanate(PEITC)on apoptosis of breast cancer cell line MCF-7.Methods After treating of MCF-7cells with different concentrations of phenethyl isothiocyanate,we used MTS assay to investigate the inhibition ratio of phenethyl isothiocyanate on MCF-7cells,flow cytometry to investigate the effect of 10and 20 μmol.L-1 phenethyl isothiocyanate on apoptosis,cell cycle and reactive oxygen.Results The phenethyl isothiocyanate could inhibit MCF-7cells proliferation.After the treatment with phenethyl isothiocyanate 72hlater,the IC50of phenethyl isothiocyanate was 15.2μmol.L-1;Affer treated with 10and 20μmol.L-1 phenethyl isothiocyanate for 24h,MCF-7cells,appeared the apoptosis,cell cycle change and oxidative damage.Conclusion Phenethyl isothiocyanate could promote the apoptosis of MCF-7cells and affect the cell cycle.Its mechanism may involve the oxidative damage of tumor cells.
