穿琥宁抑制流感病毒诱导狗肾传代细胞凋亡

目的 :探讨穿琥宁对流感病毒诱导狗肾传代细胞 (MDCK)的影响。方法 :采用观察细胞形态学改变 ,计算细胞凋亡的百分数及流式细胞仪 (FCM )测定法。结果 :病毒感染组细胞在光镜下可见细胞核产生凋亡的形态变化 ,细胞凋亡百分率 (凋亡指数AI)高于正常细胞 (P <0 .0 1) ,且A/南昌 / 692 / 97(H1 N1 )高于B/南昌 / 495 / 97毒株 (P <0 .0 1) ,说明流感病毒可诱导细胞凋亡 ,而H1 N1 型株比B型株强 ;H1 N1 型和B型流感病毒感染细胞组的AI明显高于穿琥宁与流感病毒同时作用的细胞组(P <0 .0 1)。结论 :穿琥宁可抵抗流感病毒诱导细胞凋亡 ,其抑制效率类似利巴韦林。

Inhibitory effects of Chuanhuning on the apoptosis induced by influenza virus in MDCK cells

OBJECTIVE To study the inhibitory effects of chuanhuning on the apoptosis induced by influenza virus in MDCK (Madin darby canine kidney) cells.METHODS Apoptotic cells were determined by observing the morphological changes under microscopy, calculating the percentage of apoptotic cells by microscopy analysis and flow cytometry analysis.RESULTS The apoptotic morphologic characteristics of nuclear were observed in MDCK cells infected with influenza virus by microscopic analysis. The percentage of apoptotic cells (apoptotic index, AI) in virus infected group was significantly higher than that of the control group ( P < 0.01 ). In addition the apoptosis-induced activity of influenza virus A/NanChang/692/97(H 1 N 1 ) strain was more potent than that of B/NanChang/495/97 strain ( P < 0.01 ). It indicated that influenza virus might induce apoptosis of MDCK cells and the apoptosis induced activity of the H 1 N 1 strain was higher than that of the B strain, chuanhuning was found to significantly ( P < 0.01 ) inhibit the apoptosis induced by influenza virus. CONCLUSIONS The results of the present study suggested that chuanhuning could resist against the apoptosis induced by influenza virus with the inhibitory rate comparable to that obtained by ribavirin.
