摘    要:①目的 探讨脆性部位与原癌基因在乙型病毒性肝炎(乙肝)病人中的相关性及其与原发性肝癌发生的关系。②方法 用低叶酸、低小牛血清和较高pH值培养方法,常规外周血染色体制片,对20例乙肝病人(病人组)和20例健康正常人(对照组)进行染色体脆性部位分析。③结果 病人组检出31种脆性部位,主要分布于A,B,C组染色体,其表达率为8.94%,与正常对照组(4.05%)比较差异有显著性(u=5.83,P〈0.0

Abstract:Objective To study the relationship between the fragile site and proto oncogene in the hepatitis B patients and its influence on the emergence mechanism of primary hepatocellular carcinoma(PHC). Methods A peripheral blood preparation of chromosome was made routinely. The culture of cells was done in low concentration of serum, folate and higher pH medium. 20 patients with hepatitis B as a group and healthy persons as control group. The fragile sites of the 2 groups were exactly located and analyzed. Results The fragile sites of the hepatitis B patients group were determined as 31 species mainly distributed on the chromosomes of A, B, C group. The expression rate of the fragile site (8.94%) was higher than that (4.05%) of the control. There was a striking difference between them ( u=5.83,P <0.001 ). Among the fragile sites there were 12 which overlaped the proto oncogene sites and the superposition frequency was 38.7%. Fra 2q21 overlapped two proto oucogene sites, fra 11q13 overlapped five proto oncogene site. Conclusion In hepatitis B patients, the overlap of the fragile site with the proto oncogene site can easily lead to HBV DNA integrate and play an important role in inducing PHC by activating proto oncogene.
