肾移植受者霉酚酸临床药代动力学研究

摘    要:目的研究肾移植受者的霉酚酸(MPA)药代动力学。方法 采用高效液相色谱法监测19名肾移植受者术后多剂量口服霉酚酸酯(MMF)后的MPA血药浓度,采用非线性混合效应模型法计算多剂量口服达稳态的MPA药代动力学参数。结果 MPA达峰时间Tmax为1.0 h,峰浓度Cmax为8.3μg/ml,谷浓度Cmin为2.6μg/ml,药时曲线下面积AUC0~12 h为44.9μg.h/ml。MPA药动学参数群体值CL=27.7 L/h,Vd=85.3 L,个体间变异分别为22.0%2、5.7%。结论本研究建立的群体药动学模型可较准确地描述MPA在肾移植受者的药动学特征,其预测能力尚需进一步评估。
