红景天水煎剂和红景天苷悬浊液对兔股动脉粥样斑块稳定性的影响

摘    要:目的比较在相同红景天苷浓度的条件下,红景天水煎剂和红景天苷悬浊液对于兔动脉粥样斑块稳定性的影响,并探讨红景天苷对粥样斑块的药理作用,以及红景天其他成分的药理效应。方法将30只新西兰兔以高脂饮食和股动脉球囊扩张法建立动脉粥样硬化模型,随机分为空白对照组、红景天组、红景天苷组。测定用药前后血脂变化,药物干预后血清基质金属蛋白酶-3(MMP-3)水平、粥样斑块的组织病理学变化,以及免疫组化检测粥样斑块的CD34、血管内皮细胞生长因子(VEGF)和MMP-3、胶原、仅平滑肌肌动蛋白、增殖细胞核抗原(PCNA)、单核细胞趋化蛋白-1(MCP-1)、糜酶(Chymase)的染色阳性面积。结果红景天组血清LDL-C、血清总胆固醇和血清MMP-3水平与空白对照组比较显著降低(P〈0.05,P〈0.01),但对血清三酰甘油无明显降低作用;红景天苷组对血脂和血清MMP-3的影响不明显。免疫组化检测显示,两药物干预组粥样斑块的CD34、VEGF、MMP-3、胶原、α平滑肌肌动蛋白和Chymase抗体的阳性面积都较空白对照组显著减少(P〈0.05,P〈0.01),其中红景天苷组降低Chymase的程度明显高于红景天组(P〈0.01),而降低d平滑肌肌动蛋白抗体的程度不如红景天组(P〈0.05)。此外,红景天苷组还可显著降低斑块内MCP-1、PC-NA水平,而红景天组降低不明显。结论红景天对于粥样斑块的部分药理作用是通过红景天苷来实现的,包括减少粥样斑块内新生血管和VEGF含量,抑制斑块内胶原生成和MMP-3活性;同时对血脂尤其是对LDL-C的作用,对平滑肌增殖和转化的影响,以及对MCP-1和Chymase的作用两者有所区别,表明有一些作用受到了红景天所含其他成分的影响。

Effects of Rhodiola Rosea Decoction and Salidroside Suspension on Plaque Stability in Arteriosclerosis Rabbits

Abstract:Objective To investigate and compare the effects of Rhodiola rosea decoction and Salidroside suspension with the same concentration of Salidroside on the arterioselerosis rabbit, and to analyze the effects on the plaque of Salidroside by itself and other ingredients contained in Rhodiola rasea. Methods Arteiosclerosis rabbit model was established with high-fat diet and angioplasty. Then 30 Newsealand Rabbits were divided into control group, Rhediola rosea decoction group and Salidroside suspension group. The blood lipid, serum MMP-3 and pathological histology changes were observed. CD34, VEGF, MMP-3, collagen, α-SMA, PCNA, MCP-1 and chymase positive stained area in the arteriosclerotic plaque were detected by immunohistochemistry techniques. Results Rhodiola rosea decoction could decrease serum total cholesterol, LDL-C and MMP-3(P 〈0.05 or P 〈0.01 ), but couldn't decrease serum triglyceride. Salidroside suspension had little effect on blood lipid and serum MMP-3. Two medicines could both decrease CD34, VEGF, MMP-3, chymase, α-SMA and collagen positive stained area in the arteriosclerotic plaque( P 〈0.05, P 〈0.01). Opossite chymase declined more significandy in Salidroside suspension group( P 〈0.01 ), and α-SMA declined more significantly in Rbodiola rosea decoction group(P 〈0.05). Furthermore, Salidroside suspension could decrease MCP-1 and PCNA positive stained area in the plaque, but Rhodiola rosea decoction could not. Conclusion Rhodiola rosea inhibits angiogenesis, VEGF production, collagen secretion and MMP-3 activity mainly by Sali-droside, which suppresses proliferation and differentiation of VSMC, MCP-1 activity, chymase depositon and blood lipid, especially LDL-C.
