摘    要:目的探讨特应性皮炎(atopic dermatitis,AD)儿童患者与正常儿童肠道菌群之间的相互作用情况。方法根据纳入标准选取35例特应性皮炎儿童患者为AD组(AD组),27例健康儿童作为对照组(N组)。应用Illumina Miseq测序平台对研究对象粪便细菌的16SrRNA基因V3-V4区进行高通量测序,将测序结果进行Network网络分析。结果(1)共现性网络比较显示:在门、属水平,AD组与N组以共有菌群为主,两组门水平共有物种5门,分别是p_Firmicutes(厚壁菌门)、p_Bacteroidetes(拟杆菌门)、p_Proteobacteria(变形菌门)、p_Actinobacteria(放线菌门)、p_Verrucomicrobia(疣微菌门);(2)物种相关性分析:g_Bacteroides(拟杆菌属)与g_Parabacteroides(副拟杆菌属)存在最大正相关(r=0.56,P<0.001),与g_unclassified_f_Peptostreptococcaceae(r=-0.53,P<0.001)、g_Intestinibacter(r=-0.49,P<0.001)、g_Bifidobacterium(双歧杆菌属)(r=-0.46,P<0.001)负相关相对较大。结论推测g_Bacteroidetes(拟杆菌属)与其具有正、负相关作用的菌群之间相互作用导致AD的发生,具体作用机制待进一步深入研究。

Abstract:Objective To explore the interaction of the different intestinal flora between Atopic dermatitis(AD)children and normal children.Methods According to the inclusion criteria,35 children with atopic dermatitis were selected as the case group(AD group),and 27 healthy children as the normal group(N group).The Illumina MiSeq sequencing platform was applied to high-through put sequencing methods of the 16 SrRNA gene V3-V4 region of the fecal bacteria of the study object,the sequencing results were analyzed by network analysis.Results(1)The comparison of co-occurrence networks showed that:at the level of phylum and genus,AD group and N group were dominated by common flora,and there were 5 phylum in the two groups,respectively p_Firmicutes、p_Bacteroidetes、p_Proteobacteria、p_Actinobacteria、p_Verrucomicrobia.(2)Species correlation analysis:The g_Bacteroides had the largest positive correlation with g_Parabacteroides(r=0.56,P<0.001),and a relatively large negative correlation with g_unclassified_f_Peptostreptococcaceae(r=-0.53,P<0.001)、g_Intestinibacter(r=-0.49,P<0.001)、g_Bifidobacterium(r=-0.46,P<0.001).Conclusion It is speculated that the interaction between the g_Bacteroides and its positive and negative related flora will lead to the occurrence of AD,and the specific mechanism needs to be further studied.
