运动训练及大豆多肽对高脂饮食诱导大鼠脂肪肝形成的干预作用

摘    要:目的:探讨运动训练和补充大豆多肽对高脂饮食诱导非酒精性脂肪肝(NAFLD)形成的干预作用。方法:雄性SD大鼠随机分为普通饮食组(C组)、高脂饮食组(HF组)、高脂饮食+运动组(HE组)、高脂饮食+大豆多肽组(HS组)。HE组大鼠进行10周、每周6次、每次60min的无负重游泳运动。HS组按500 mg/kgBW/d的剂量用大豆多肽(SPH)灌胃。10周后测定大鼠体重、肝重、血清和肝组织TG、TC含量及肝脏AMPK基因表达量;HE染色进行肝细胞形态学观察。结果:(1)与C组相比,HF组大鼠肝重、肝指数、血清及肝脏TG、TC含量显著增加(P<0.05),肝脏AMPK mRNA显著下降(P<0.05);(2)与HF组相比,HE组大鼠血清TG、TC含量显著降低(P<0.01),肝脏AMPK mRNA表达显著上升(P<0.05);(3)与HF组相比,HS组大鼠肝重、血清和肝脏TG、TC含量显著下降(P<0.05),肝脏AMPK mRNA略有上升(P>0.05);(4)光镜下HF大鼠肝细胞呈中、重度脂肪变,HE组、HS组大鼠肝组织细胞正常。结论:10周高脂饮食可诱导大鼠NAFLD形成,运动训练或补充SPH可明显预防高脂饮食诱导大鼠NAFLD形成。

Abstract:Objective To study the effect of exercise and soy protein hydrolyzate(SPH)on the formation of nonalcoholic fatty liver disease(NAFLD)induced by high-fat diet.Methods SD rats were divided randomly into normal diet group(C),high-fat diet group(HF),high-fat diet plus swimming group(HE),and high-fat diet plus SPH group(HS).Rats in the group HE underwent unloaded swimming for 10 weeks(60min/day,6days/week).Rats in the group HS were treated intragastrically with SPH,500mg/kgBW·d.BW(body weight).LW(liver weight),TG,TC and AMPK mRNA in liver or serum were detected,and the morphological changes in the liver tissues were observed with microscope at the end of experiment.Results(1)Compared with group C,LW,LW/BW,TG,TC in liver and serum increased significantly(P < 0.05),the AMPKmRNA in liver decreased(P < 0.05)significantly in group HF;(2)Compared with group HF,TG and TC in serum decreased markedly(P < 0.01),and AMPK mRNA in liver increased significantly(P < 0.05)in group HE;(3)Compared with group HF,LW,TG and TC in liver and serum decreased significantly(P < 0.05),and AMPK mRNA in liver increased slightly(P > 0.05)in group HS;(4)More or severe steatosis hypatocytes was found under microscope in group HF,the histology of tissues in groups HE and HS was restored to nearly normal.Conclusion Feeding high-fat diets for ten weeks could induce the formation of NAFLD in rats.Exercise or supplementation of SPH could effectively prevent the formation of NAFLD induced by high-fat diet in the rats.
