红细胞生成素对大鼠脑缺血再灌注后神经元保护作用

摘    要:目的 观察促红细胞生成素(erythropoietin,EPO)对大鼠脑缺血再灌注后的梗死体积及神经功能缺损评分的影响。方法 线栓法复制大鼠左侧中动脉缺血再灌注模型,缺血2h,于再灌注开始时经腹腔注入重组人促红细胞生成素(3000U/kg),再灌注后2h,12h,24h,48h,72h,7d及10d测定大鼠神经功能缺损评分后,断头取脑,用2%氯化三苯基四氮唑(TTC)标记梗死体积。结果 对照组缺血再灌注后2h已经出现梗死灶,随着时间的延长,梗死灶体积逐渐增大,24h时梗死体积最大。EPO治疗组与相应时间点对照组相比,梗死灶体积明显缩小(P〈0.05),神经功能缺损评分明显减少(P〈0.05)。结论 24h时达最大,EPO能使梗死灶体积明显缩小、神经功能缺损评分明显减少,对损伤神经元有保护作用。

Abstract:Objective To investigate the effects of erythropoietin on infarction volume and the scores of neurological deficits after focal brain ischemia and reperfusion in rat. Methods The model of focal brain ischemia was made by occluding middle cerebral artery(MCA) for 2 h and reperfusion for 2,6,12,48,72 h,7 d and 10 d.The rats received rhEPO(3 000 U/kg of body weight)or saline i.p before the beginning of reperfusion.The score of neurological deficits were determined at the time points.The infarction volume was marked by 2%triphenyltetrazolium chloride(TTC) at 2,6,12,48 and 72 h,7 d and 10 d. Results The infarction was detected at 2 h after focal brain ischemia and reperfusion.With prolonging of time,the infarction volume became more and more large.The infarction volume peaked at 24 h after ischemia and reperfusion.Compared with control group,the infarction volumes were significantly smaller at 6,12,48 and 72 h,7 d and 10 d in EPO group.Conclusion The results indicate that EPO has protective effect on neuron injury in rats with focal brain ischemia and reperfusion.
