智脑胶囊对阿尔茨海默病模型大鼠脑组织BDNF及其TrkB影响的实验研究

摘    要:目的:观察智脑胶囊对实验性阿尔茨海默病(AD)模型大鼠脑组织脑源性神经营养因子(BDNF)及其酪氨酸蛋白激酶受体B(TrkB)的影响,探讨智脑胶囊治疗AD的作用机制。方法:雄性Wistar大鼠,随机分为7组,即正常对照组、假手术组、AD模型组、石杉碱甲组、智脑胶囊低、中、高剂量组。参照《大鼠脑立体定位图谱》,选双侧Meynert基底核为注射点,以鹅膏蕈氨酸(IBO)毁损双侧Meynert基底核,制作实验性AD大鼠模型。用ELISA酶联免疫法检测脑组织BDNF及其受体TrkB的含量,并观察其在大鼠脑组织皮层和海马不同部位含量的变化。结果:智脑胶囊能提高模型大鼠脑组织(皮层和海马)BDNF及其受体TrkB的含量,与模型组比较,差异有显著性(P0.05)。其中智脑胶囊高、中剂量组和石杉碱甲组的提高作用优于智脑胶囊低剂量组。结论:智脑胶囊促进大鼠脑组织皮层和海马神经元合成BDNF及其受体TrkB表达增加是其改善学习记忆的重要作用机制。
