骨吸收抑制剂对猕猴皮质骨形态计量学的影响

摘    要:目的评估骨吸收抑制剂对猕猴皮质骨骨形态计量学指标的影响。方法 20只雌性猕猴(18~22岁)按体重随机分为5组:对照组(CNT)、低剂量依降钙素组[0.5 U/(kg·d),ELL]、高剂量依降钙素组[5 U/(kg·d),ELH]、低剂量阿仑膦酸钠盐组[10μg/(kg·d),ALL]和高剂量阿仑膦酸钠盐组[100μg/(kg·d),ALH]。皮下注射给药29周,每周2次。处死前静脉注射钙黄绿素,处死后取左侧股骨中段横截面进行骨形态计量学检测。结果 5组左侧股骨截面总面积(Tt.Ar)、皮质骨面积(Ct.Ar)、髓腔面积(Ma.Ar)、类骨质周长(O.Pm)和类骨质宽度(O.Wi)的差异均无统计学意义(均P0.05)。用药各组皮质骨骨吸收腔周长(E.Pm)均较对照组明显降低(均P0.05)。ALH组骨矿沉积率(MAR.)低于对照组(P=0.049)。ELL、ALL、ALH组修正后的沉积速率(AJ.AR)低于对照组(P=0.040、0.018、0.09)。各组骨矿化延迟时间(Mlt)、修正后的骨矿化延迟时间(O.Wi/AJ.AR.)和骨形成率(BFR)间差异无统计学意义(P=0.171、0.110、0.122)。结论降钙素及阿仑膦酸钠通过抑制皮质骨破骨细胞功能抑制皮质骨的骨重建,二膦酸盐作用强于降钙素。降钙素及阿仑膦酸钠能不同程度地影响MAR,但对Mlt及总BFR影响不大。

Effect of antiresorptive drugs on cortical bone of cynomolgus monkeys by histomorphometrical analysis

Abstract:Objective To evaluate the effect of eel calcitonin ( elcatonin ) and alendronate on cortical bone of cynomolgus monkeys by histomorphometrical methods .Methods Twenty female cynomolgus monkeys ( Macaca fascicu-laris), aged 18-22 years, were allocated into five groups:control (CNT, n=4), low-dose elcatonin group (0.5 U/kg;ELL, n=4), high-dose elcatonin group (5 U/kg;ELH, n=4), low-dose alendronate group (10 Ug/kg;ALL, n=4) and high-dose alendronate group (100 Ug/kg;ALH, n=4).All animals were given subcutaneous injections twice a week for 29 weeks and received double bone fluorochrome labeling prior to sacrifice .At the end of the experiment , the left fe-mur of monkeys were harvest for bone histomorphometrical evaluation .Results Tt.Ar, Ct.Ar, Ma.Ar, O.Pm, and O.Wi in elcatonin-treated groups and alendronate-treated groups were similar to CNT group ( P>0.05 ) .E.Pm in medi-cation groups were significantly lower ( P<0.01 ) than that in CNT group , and E.Pm in ALH group was much lower than that in ELL group ( P=0.027 ) . CNT group had a higher AJ.AR than the ELL and alendronate-treated groups ( P=0.040, 0.018, 0.09), whereas, only ALH group had a lower MAR than the other groups (P=0.049).Mlt, O.Wi/AJ.AR, BFR had no significant differences in each group (P=0.171, 0.110, 0.122).Conclusion Calcitonin and alendronate can both inhibit bone resorption by reduce the osteoclast function , and the alendronate seems has a stronger effect than calcitonin .The use of calcitonin and allendronate will affect the bone MAR in varying degrees , wherease , it has little effect on the Mlt and BFR .
