培哚普利对自发性高血压大鼠肾脏核因子-κB表达的影响

摘    要:目的 :探讨培哚普利对自发性高血压大鼠肾脏核因子 -κB表达的影响。方法 :采用自发性高血压大鼠 ( SHR)模型 ,以核因子 ( NF) -κB亚基 P65多抗为抗体 ,应用免疫组织化学染色法观察培哚普利对 SHR肾脏 NF-κB表达的影响 ,并进一步分析 NF-κB表达与肾小球内粘附分子 - 1 ( ICAM- 1 )蛋白表达、单核细胞浸润及肾脏功能和结构损害的关系。结果 :SHR肾小球 NF-κB表达较正常肾组织增高 ,培哚普利可显著降低 NF-κB表达。且 NF-κB阳性细胞数与肾小球 ICAM- 1蛋白表达、单核细胞浸润、肾脏功能和结构损害显著相关。结论 :培哚普利对 SHR肾脏保护作用可能通过抑制血管紧张素 引起的 NF-κB活化 ,下调受 NF-κB调控的 ICAM- 1表达 ,减轻单核细胞浸润而实现。

Effects of perindopril on renal NF-Κb expression in spontaneous hypertensive rats

Abstract:Objective: To investigate the effects of perindopril on renal NF-κB expression in spontaneous hypertensive rats. Methods: With the model of spontaneous hypertensive rats(SHR),the effects of perindopril on renal NF-κB P65 expression in SHR were studied with immunocytochemical techniques using NF-κB P65 as antibody. At the same time,the relationship between glomerular expression of NF-κB and ICAM-1 expression,macrophage accumulation in SHR was examined. Results:The expression of NF-κB in glomerulus of SHR was higher than in the normal control,while perindopril could decrease the expression of NF-κB in SHR.The number of positive cells expressing NF-κB in glomerulus of SHR showed a strong correlation with ICAM-1 expression,macrophage accumulation and urine protein excretion,respectively. Conclusion: Perindopril might protect hypertensive renal tissue by inhibiting angiotensin-stimulated NF-κB activation,therefore downregulating ICAM-1 expression and macrophage infiltration which might play a key role in hypertensive nephropathy.
