组织因子途径在灯盏花素肺部给药抗急性肺损伤的作用研究

摘    要:目的从TF途径研究灯盏花素肺部给药抗内毒素(lipopolysaccharide,LPS)致大鼠ALI的作用.方法健康雄性SD大鼠120只随机分成五组(n=24):正常组、生理盐水组、灯盏花素组、LPS组、灯盏花素+LPS组;分别观察2、6、12 h大鼠的血浆和支气管肺泡灌洗液(BALF)中TF和TFPI的含量,计算TF/TFPI比值.结果与LPS组相比,灯盏花素+LPS组大鼠血浆和BALF中TF含量明显下降(P〈0.05),血浆和BALF中TFPI含量明显降低(P〈0.05);与正常对照组和生理盐水组相比,LPS组各时间点大鼠血浆和BALF中的TF/TFPI比值显著增高(P〈0.05),灯盏花素+LPS组与LPS组相比,血浆和BALF中TF/TFPI比值明显降低(P〈0.05),但是仍高于正常对照组和生理盐水组.结论灯盏花素肺部给药具有抗ALI的作用,其作用与抑制TF和TFPI的生成有关.

Abstract:Objective To explore the therapeutic effect of pulmonary delivery of breviscapine on rats with acute lung injury induced by lipopolysaccharide from tissue factor pathway.Methods Healthy male adult Sprague-Dawley rats were randomly divided into five groups(n=24 in each group):normal control group,saline group,breviscapine group,LPS group,and breviscapine+LPS group.At 2 h,6 h and 12 h after treatment,the concentrations of TF and TFPI in BALF and plasma of rats in each group were measured and the TF/TFPI ratio was calculated.Results Compared with LPS group,the concentrations of TF and TFPI were decreased significantly(P〈0.05).Compared with normal control group and saline group,TF/TFPI ratio in BALF and plasma in LPS group increased significantly(P〈0.05).TF/TFPI ratio in BALF and plasma of rats in breviscapine+LPS group was significantly lower than that in LPS group,but still higher than that in normal control group and saline group(P〈0.05).Conclusions(1)Pulmonary delivery of Breviscapine has anti-ALI effects.(2)The anti-ALI effects of pulmonary delivery of Breviscapine are related to inhibiting the formation of TF and TFPI.
