摘    要:本研究探讨stathmin1基因和蛋白在急性白血病初治、复发和缓解患者中的表达及其意义.应用FQ-PCR和Western blot检测25例正常人和76例急性白血病患者外周血细胞中stathmin1 mRNA和stathmin1蛋白的表达水平.结果显示,在正常人体内检测不出stathmin1蛋白表达,其mRNA在正常人体内则低水平表达.急性白血病初治患者stathmin1 mRNA表达高于正常人(P<0.05),急性白血病复发者stathmin1 mRNA表达显著高于初治者(P<0.05),急性髓系白血病与急性淋巴细胞白血病之间stathmin1 mRNA表达无显著差别(P>0.05).急性白血病初治患者stathmin1蛋白表达的阳性率为89%,stathmin1蛋白在急性白血病患者缓解后表达明显下降或不表达,急性白血病复发者stathmin1蛋白表达与初治者无显著差别(P>0.05),急性髓系白血病与急性淋巴细胞白血病之间stathmin1蛋白表达无显著差别(P>0.05).结论:stathmin1蛋白和mRNA在急性白血病初治及复发患者体内高表达,而在缓解后体内仍有弱表达的患者,具有一定的复发风险,因此stathmin1可能成为判断微残留白血病的一个新的生物学指标.

Abstract:This study was aimed to investigate the expression of stathminl mRNA and stathminl protein in de novo patients with acute leukemia (AL) ,relapsed patients with AL and complete remission patients with AL, and its clinical significance. The expression of stathminl mRNA and stathminl protein in peripheral blood samples from 76 cases of AL and 25 healthy persons were examined by fluorescent quantitative PCR(FQ-PCR) and Western blot, respectively. The results showed that the stathminl protein expression could not be detected in healthy persons, only the low level of its mRNA could be observed in them. The stathminl mRNA expression level in de novo AL patients was higher than that in healthy persons ( P 〈 0.05 ), the stathminl mRNA expression level in relapsed patients with AL was higher than that in de novo patients ( P 〈 0.05 ), and there was no signiticant difference of stathminl mRNA expression between patients with AML and patients with ALL. The positive rate of stathrninl protein expression in de novo patients with AL was 89%, while it obviously decreased or did not express in complete remission patients with AL. The stathminl protein expression in relapsed patients with AL did not display significant difference as compared with that in de novo patients ( P 〉 0.05 ). There was no significant difference in stathminl protein expression between patients with AML and patients with ALL (P 〉 0.05). It is concluded that stathminl protein and mRNA are overexpressed in de novo patients and relapsed patients, and lowly expressed in complete remission patients. Therefore, the stathminl may be a new biological marker for evaluation of minimal residual disease.
