提高大鼠实验性肝硬变模型存活率的研究

摘    要:目的探讨提高大鼠肝硬变模型制作存活率的方法.方法将120只雄性SD大鼠随机分为3组:实验1组(40只):CCL4+10%乙醇,10%乙醇作为唯一饮料;实验2组(40只):CCL4+30%乙醇,在基础饲料添加20%猪油、0.5%胆固醇连续喂养2周,30%乙醇在进行CCL4注射前用于灌胃,用高压消毒的水作为饮料;正常对照组(40只):按常规饲养.观察两种方法诱导大鼠实验性肝硬变模型的存活率和肝组织的病理变化.结果实验1组(40只)SD大鼠肝硬变模型诱导8周共死亡15只,存活率为62.5%;实验2组(40只)8周共死亡2只,存活率为95%(P<0.05).结论在进行CCL4皮下注射前用乙醇作间歇性灌胃可以明显提高大鼠肝硬变模型的存活率。

Abstract:Objective To investigate a method to increase the survival rate of cirrhotic model in rats. Methods 120 male SD rats were randomly divided into three groups: The first group (n=40): CCL+10% alcohol, 4 alcohol solution (10%) as sole drinking fluid 1]. The second group (n =40): CCL4+30% alcohol, the rats were fed with basic diet, which was added 20% lard and 0.5% cholesterol for 2 weeks. 30% alcohol is used intragastrically before the injection of distilled water as drinking water; the control group (n=40) was conventionally treated. The survival rate and liver cell change in two ways were observed. Results After being induced with above means for 8 weeks, the SD rat survival rate of the first group is 62.5%, there are 15 rats dead in all; the SD rat survival rate of the second group is 95% (P < 0.05), there are 2 rats dead in all. Conclusion The cirrhotic model in rats were induced by using alcohol interval intragastrically before CCL4 injection showed a high survival rate.
