脊髓损伤后毛细血管新生与血管内皮生长因子mRNA的表达

摘    要:目的探讨脊髓损伤后毛细血管新生与血管内皮生长因子(VEGF)mRNA的表达。方法采用改良Allen's重量打击法大鼠急性脊髓损伤模型,用马松三色染色法观察脊髓损伤后毛细血管的新生情况,原位杂交法观察急性损伤后大鼠脊髓VEGF mRNA表达的动态变化。结果脊髓损伤后引起脊髓组织中血管密度和血管分布改变,脊髓急性损伤后损伤局部出现短暂的血管新生过程。同时,原位杂交结果显示VEGFmRNA的表达上调,表达时间与血管变化的时间重叠。除了时间上的相互关联外,脊髓损伤后毛细血管新生与VEGF mRNA的表达在空间分布上是一致的。结论脊髓急性损伤可上调VEGFmRNA在脊髓血管内皮细胞、胶质细胞和巨噬细胞内的表达,有短暂的血管新生过程,损伤组织中VEGFmRNA的表达与其毛细血管新生的调节密切相关。

Abstract:Objective To investigate the response of the vasculature and time course of the expression of mRNA for the angiogenic vascular endothelial growth factor(VEGF) in rats spinal cord following acute injury.Methods The spinal cord injury model was made by modulated Allen's weight drop method.The response of the vasculature was determined by using Masson trichrome,and the expression of VEGF mRNA was assayed by using in situ hybridization.Results Lesion models induced changes in the density and the distribution pattern of blood vessels: a transient local sprouting of vessels was induced upon traumatic spinal cord injury.In situ hybridization showed that there was an increase of VEGF mRNA anticipated and the overlap with the vascular responses in lesion models.In addition to the temporal correlation of VEGF expression and vascular sprouting,there was a clear correlation in the spatial distribution patterns. Upon lesion in the adult animal,VEGF was mainly detectable at the border of the lesion area, where the transient increase in vascular density could be observed.Conclusion Lesion models can induce changes in the density and the distribution pattern of blood vessels and an increase of the expressions of VEGF mRNA.Results strongly indicate that VEGF is crucially involved in mediating vascular changes following traumatic spinal cord injury in the central nervous system.
