摘    要:目的了解新诊断2型糖尿病外周血单个核细胞中氨基末端激酶(JNK)(Thr^183/Tyr^185)活性变化及其影响因素。方法招募正常糖耐量健康志愿者60名(NGT组),新诊断2型糖尿病患者60例(NT2DM组)。应用western blot法检测外周血单个核细胞中JNK(Thr^183/Tyr^185)磷酸化水平。结果NT2DM组JNK(Thr^183/Tyr^185)磷酸化水平显著高于NGT组,差异有统计学意义(P〈0.01)。相关性分析显示,JNK(Thr^183/Tyr^185)磷酸化水平与体重指数(BMI)、空腹血糖(FBG)、低密度脂蛋白胆固醇(LDL—C)呈正相关(P〈0.05),在剔除年龄、性别等相关影响因素之后,空腹血糖仍与JNK(Thr^183/Tyr^185)磷酸化水平独立相关。结论JNK通路过度激活可能增加了患糖尿病的风险。

Abstract:Objective To explore the change of JNK (Thr^183/Tyr^185) activity in the peripheral blood mono- nuclear cells in the newly diagnosed type 2 diabetes patients. Methods A total of 60 healthy volunteers with normal glucose tolerance (NGT group), 60 newly diagnosed type 2 diabetes patients (NT2DM group) were recruited in our study. JNK (Thr^183/Tyr^185 ) phosphorylation levels were deleeted in the peripheral blood mononuclear cells by western blot method. Results JNK ( Thr^183/Tyr^185 ) phosphorylation levels in NT2DM group were significantly higher than that in NGT group (P 〈 0. 01 ). The correlation analysis showed that JNK (Thr^183/Tyr^185) phosphorylation levels were positively associated with body mass index, fasting blood glucose and low-density cholesterol ( P 〈 0. 05 ). Under the situation of excluding the related factors such age, gender and so on, JNK ( Thr^183/Tyr^185 ) phosphorylation levels were still independently associated with fasting blood glucose ( P 〈 0. 05). Conclusion JNK pathway over-activation may increase the risk of diabetes.
