鹿线粒体DNA的种属特异性片段研究

摘    要:目的:寻找鹿线粒体DNA上区别于常见伪充鹿类药材来源动物的种属特异性片段。方法:根据对不同产地梅花鹿、马鹿、白唇鹿线粒体DNA进行PCR扩增和序列测定,并与亲缘关系较近的伪充药材来源动物线粒体DNA同位置序列比较,找到鹿的特征片段,经过实际检测,筛选出鹿的种属特异性片段。结果:该种属特异性片段作为PCR引物能对鹿DNA进行扩增,而不能扩增其它常见伪充药材来源动物的DNA。结论:该片段能从基因型特征上将鹿和伪充药材来源动物区分开来,而且准确可靠。

Abstract:OBJECTIVE:To find the species distinctive segment of mtDNA which can be used to distinguish the deer from those animals always used to make false medicine of deer.METHODS:The mtDNA of sika, red deer, sambar and white lipped deer from different areas were amplified by PCR and sequenced. We found the characteristic segments of deer after comparing them with the mtDNA of animals of the same clan from which the false medicine were made. We selected one as the species distinctive segment after the practical amplification. RESULTS:When the segment was used as PCR primer, the DNA of deer could be amplified but that of animals always used to make false medicine of deer could not. CONCLUSION:This segment can distinguish the deer from those animals always used to make false medicine of deer from the genotype and it is also accurate.
