摘    要:目的研究髓鞘碱性蛋白(MBP)对实验性脑出血大鼠神经保护作用机制。方法选择SD大鼠60只,脑出血造模成功后随机分为MBP组、磷酸盐缓冲盐水(PBS)组、卵白蛋白(OVA)组,每组30只。每天应用Longa五级评分法进行神经功能缺损评分,不同亚组按相应时间点取材,分别测定出血周围脑组织胶质纤维酸性蛋白(GFAP)和脑源性神经营养因子(BDNF)的免疫组化评分。取脾组织制作匀浆,反转录聚合酶链式反应(RT-PCR)测定Foxp3与β-actin的相对密度值。结果 MBP组在对应时间点神经功能缺损评分及GFAP和BDNF的表达均明显高于OVA组和PBS组(P0.05或0.01);OVA组与PBS组比较均无显著性差异。Foxp3mRNA在大鼠脾脏中的表达MBP组均明显低于OVA组和PBS组(P均0.01),OVA组与PBS组比较无显著性差异。结论 MBP对实验性脑出血后大鼠的行为学缺损有治疗作用,可减轻症状,上调BDNF、GFAP蛋白表达水平,降低Foxp3 mRNA的表达;对血肿周边神经元产生保护作用,促进中枢神经系统损伤的恢复。

Study of the mechanism of action of MBP for promoting rehabi il tation in the rats with intracerebral hemorrhage

Abstract:Objectiev It is to study the mechanism of nerve protection of myelin basic protein ( MBP) in the rats with ex-perimental intracerebral hemorrhage .Methods 60 SD rats were selected and were randomly divided into MBP group , phos-phate buffered solution ( PBS) group and ovalbumin ( OVA) group after intracerebral hemorrhage models were successfully es-tablished, each group had 30 rats.Neurological deficit scores were evaluated every day by Longa 5 degree scores.All the mod-el rats were drawn the materials on the corresponding time .The expression of GFAP and BDNF in the brain tissue was detected with immuno-histochemistry .Homogenate was made from spleen to detect the relative light density of Foxp 3 and β-action by RT-PCR.Results Neurological deficit scores on the corresponding time and the expressions of GFAP and BDNF in MBP group were higher than those in OVA group and PBS group (P〈0.05 or 0.01), but there was no significant difference in the indexes above between OVA group and PBS group .The expression of Foxp3 mRNA in the rats’ spleen tissue was obviously lower than that in OVA group and PBS group , but no significant difference was found between the latter two groups .Conclu-sion MBP is beneficial to improving the behavioral deficit in experimental intracerebral hemorrhage rats , which can relieve symptoms, increase the expression of BDNF and GFAP , decrease the expression of Foxp3mRNA,thus to protect the nerve cells around the hematoma and promote the recovery of central nervous system injury .
