青蒿素对糖尿病大鼠肾组织AP-1的DNA结合活性升高的抑制作用

摘    要:目的探讨青蒿素对实验糖尿病大鼠肾组织转录因子活化蛋白-1(AP-1)的DNA结合活性升高的抑制作用。方法将75只实验大鼠分成正常对照组(A组)、糖尿病非治疗组(B组)及青蒿素治疗组(C组),B、C组应用腹腔单剂量注射链脲佐菌素(STZ)65 mg/kg建立糖尿病大鼠模型。C组给予腹腔注射青蒿素300 mg/(kg·d)。于实验进行到第4周时处死各组大鼠,取部分右肾皮质,应用电泳迁移率改变分析(EMSA)法观察青蒿素对实验糖尿病大鼠肾组织AP-1的DNA结合活性的影响,并制备病理切片以观察肾组织的病理学变化。结果 B组大鼠肾细胞AP-1的DNA结合活性较A组显著上升,C组大鼠对肾细胞AP-1的DNA结合活性有显著抑制作用,并显著改善糖尿病大鼠肾脏组织的病理变化。结论青蒿素通过下调糖尿病大鼠肾组织AP-1的DNA结合活性上调而改善糖尿病大鼠肾脏组织病理改变。

Abstract:Objective It is to approach the inhibitory effect of artemisinin on the upregulation of the DNA - binding activity of AP - 1 in the kidney tissue of experimental diabetic rats. Methods 15 rats were divided into normal control rats ( group A) , STZ -induced diabetic rats (group B) , STZ- induced diabetic rats treated with artemisinin 300 m/( kg ·d) intrapcritoneally (group C). The DNA -binding activity of AP-1in the kidney tissue was detected by electrophoretic mobility shift assay (EMSA) , while renal pathologic changes were observed after 4 weeks of treatment. Results The DNA - binding activity of AP - 1 was significantly activated in renal cortices in untreated - diabetic rats after 4 weeks. The abnormal level of the DNA - binding activity of AP - 1 was partly reversed in diabetic rats treated by artemisinin. Accordingly, renal pathologic changes in diabetic rats treated with artemisinin were significantly improved. Conclusion Artemisinin has some renoprotective effects in experimental diabetic rats partly through inhibiting the DNA - binding activity of AP - 1 in renal cortices.
