间接竞争酶联免疫吸附法检测骨肽的研究

摘    要:目的:研究酶联免疫吸附法对骨肽的检测。方法:考察抗原浓度、竞争时间、二抗浓度、工作时间、显色时间等对间接竞争酶联免疫吸附结果的影响。结果:检测抗原浓度为0.25μg/ml,竞争时间30min,酶标二抗的最佳工作浓度为1∶400,二抗反应时间40min,显色时间15min。结论:本实验通过优化间接酶联免疫检测条件,建立了骨肽的间接竞争酶联免疫吸附检测方法,试验结果稳定,且精确度较好。

Abstract:Objective: To study the enzyme-linked immunosorbent assay for the detection of bone peptide. Methods: Investigated the antigen concentration, competition time, the second antibody concentration, working hours, the color time on indi - rect competitive enzyme-linked immunosorbent assay results. Results: The detection of antigen concentration was 0.25 μg/ml, competition time was 30 min, HRP secondary antibody concentration of 1 ∶400 of the best work, two anti-reaction time was 40 min, color time was 15 min. Conclusion: In this study, by optimizing the indirect enzyme-linked immunosorbent assay conditions, the establishment of bone peptide competition ELISA indirect detection methods, test results are stable, and there is better accuracy.
