中药肾炎宁对IgA肾病大鼠巨噬细胞移动抑制因子的影响

摘    要:目的探讨肾炎宁对IgA肾病大鼠肾组织巨噬细胞移动抑制因子(macrophage migration inhibitory factor,MIF)的影响。方法将Sprague—Dawley(SD)大鼠随机分为4组:对照组、模型组、西药组(贝那普利+氯沙坦)、中药组(肾炎宁),按各组相应药物剂量灌胃。于第15周末检测各组大鼠24h尿蛋白量、尿红细胞及IgA免疫荧光检测,并用免疫组织化学法检测MIF蛋白表达水平。结果中药组、西药组大鼠与模型组大鼠24h尿蛋白量比较差异有统计学意义(P〈0.01),且中药组、西药组与模型组比较IgA免疫荧光强度显著下降;中药组与模型组尿红细胞计数比较显著降低,差异有统计学意义(P〈0.01),而西药组与模型组尿红细胞计数比较差异无统计学意义(P〉0.05);中药组、西药组与模型组比较MIF表达水平显著下降,差异有统计学意义(P〈0.01)。结论肾炎宁可能通过抑制肾组织MIF表达率发挥对IgA肾病的治疗作用。

The Effect of Shenyanning on IgA Nephropathy Rats in Terms of the Expression of Macrophage Migration Inhibitory Factor
Abstract:Objective To investigate the effect of Shenyanning on the expression of macrophage migration inhibitory factor (M1F) in IgA nephropathy rats. Methods SD rats were randomly divided into 4 groups: control group, model group, western medicine group and Shenyanning group, and afforded with the medicine respectively. At the fifteenth week, 24 h urine protein quant (UPro) , the RBC morphous in urine, IgA immunofluorescence and MIF were detected with relative technique. Results In western medicine group and Shenyanning group, the levels of UPro, IgA immunofluorescence were significantly de-creased in contrast with model groups (P <0. 01) ; Compared with model group, the RBC morphous in urine in Shenyanning group were decreased significantly (P < 0. 01) , while it were not statistically significant in the western medicine group (P > 0. 05) ; In western medicine and Shenyanning groups, the levels of the expression of MIF were significantly decreased in contrast with model group (P<0. 01). Conclusion Shenyanning could treat IgA nephropathy rats effectively by suppressing the expres-sion of macrophage migration inhibitory factor.
