摘    要:目的:建立同时测定三黄片中7种有效成分——汉黄芩苷、汉黄芩素、大黄素、芦荟大黄素、大黄酸、大黄酚、大黄素甲醚含量的方法。方法:采用毛细管电泳法。内标为对乙酰氨基酚;毛细管柱为未涂层弹性融硅石英柱,有效长度为47 cm;运行缓冲液为25 mmol.L-1硼砂溶液+25 mmol.L-1十二烷基硫酸钠+4 mmol.L-1磺丁基β-环糊精+8%甲醇(pH 9.42);分离电压为14 kV;重力进样时间为5 s(高度为15 cm);检测波长为254 nm;温度25℃;湿度<70%。结果:7种有效成分检测浓度分别在2.02~20.16、1.00~10.00、1.60~16.00、1.20~12.00、2.00~20.00、7.20~72.00、1.00~10.00μg.mL-1范围内与各自峰面积积分值线性关系良好;回收率分别为100.6%、98.3%、99.9%、101.2%、99.8%、102.0%、101.8%,RSD均<3.0%。结论:本方法专属性强、结果准确可靠、重复性好,可用于三黄片的质量控制。

Abstract:OBJECTIVE: To establish a method for the content determination of seven effective component, including wogonoside, wogonin, emodin, aloe-emodin, rhein, chrysophanol, physcion, in Sanhuang tablets. METHODS: Capillary electrophoresis was adopted using paracetamol as internal standard. The separation was performed on a uncoated fused silica capillary with 47 cm effective length. 25 mmol· L^-1borax solution+25 mmol· L^-1 SDS+4 mmol. L^-1 SBE-μ-CD+8 % methanol solution (pH 9.42 ) was se- lected as the running buffer. The voltage applied was 14 kV. The sample was injected by gravity(5 s, 15 cm). The detection wave- length was 254 nm. The column temperature was 25 ~C and humidity was lower than 70%. RESULTS: The calibration curves lbr the seven components were linear in the concentration range of 2.02~20.16, 1.00~10.00, 1.60~16.00, 1.20-12.00, 2.00 ~-- 20.00, 7.20~72.00, 1.00~10.00 p.g.mL '. The average recoveries were 100.6%, 98.3%, 99.9%, 101.2%, 99.8%, 102.0%. 10t.8%, respectively, with RSD not more than 3.0%. CONCLUSION: The method is proved to be specific, accurate, reliable and well rc- peatable, and can be used for quality control of Sanhuang tablets.
