左旋四氢巴马汀在大鼠急性脑缺血再灌注时对Bcl-2、Bax蛋白表达的影响

摘    要:目的 观察左旋四氢巴马汀 (L tetrahydropalmatine ,L THP)在脑缺血再灌注时对凋亡相关基因Bcl 2、Bax蛋白表达的影响。方法 建立大鼠急性全脑缺血再灌注模型 ,采用免疫组织化学法、原位末端标记法 ,分别检测假手术组 (S组 )、缺血再灌注组 (Ⅰ组 )、L THP治疗组 (T组 )海马CA1区Bcl 2、Bax蛋白表达水平及凋亡细胞数。结果  (1)脑缺血再灌注时 ,海马CA1区Bcl 2蛋白的表达显著增加 (P<0 0 1) ,但随时间的延长 ,增加幅度逐渐减小 ;而T组Bcl 2蛋白表达于相应的时间点则明显大于I组 (P<0 0 1)。 (2 )I组各时间点Bax蛋白表达均大于S组 (P <0 0 1) ,且随时间延长增加幅度逐渐增大 ;而T组Bax蛋白表达在相应时间点则下降 (P <0 0 1)。 (3)T组的神经细胞凋亡数与Bcl 2 /Bax蛋白阳性细胞数比值的关系为负相关 :r=- 0 94 173,P <0 0 0 1。 (4)神经细胞凋亡数随再灌注时间延长而增加 ,L THP可减少细胞凋亡数。结论 在脑缺血再灌注细胞凋亡过程中 ,L THP可通过上调Bcl 2蛋白表达 ,下调Bax蛋白表达 ,减少神经元凋亡 ,对脑缺血损伤起保护作用

Abstract:Objective To investigate the effects of L Tetrahydropalmatine(L THP)on the expression of Bcl 2 and Bax after cerebral ischemia and reperfusion.Methods The model of global cerebral ischemia and reperfusion was induced.The Wister rats were randomly divided into three groups:sham operation group(S),ischemic reperfusional group(I),and L THP treated group(T).The changes of Bcl 2 and Bax protein expression were evaluated by immunohistochemical method.The number of neuronal apoptosis was accounted by Situ end labeling.Results Compared with sham operation group,the expressions of Bcl 2 and Bax positive cells were increased significantly in ischemic reperfusion group( P <0 01) and the number of neuronal apoptosis was also increased( P <0 01).But with increasing duration of reperfusion,the level of Bcl 2 gradually decreased and that of Bax was still increased.Meanwhile after L THP treatment,the expression of Bcl 2 was up regulated and that of Bax was down regulated;and the number of neuronal apoptosis was decreased( P <0 01).The number of Bcl 2/Bax positiue ceus was negatinaly correlated with that of neuronal of optosic ceus:r=-0 94173, P <0 001.Conclusion Our results indicate L THP can reduce the apoptosis through regulating Bcl 2 and Bax.
