人结肠癌与结肠腺瘤的定量蛋白质组学研究

摘    要:目的 研究结肠癌(CC)与结肠腺瘤性息肉(CAP)组织差异定量表达蛋白质谱.方法 激光捕获显微切割(LCM)技术分别获取结肠癌及结肠腺瘤性息肉组织上皮细胞,应用同位素标记相对和绝对定量(iTRAQ)及纳米液相色谱-串联质谱(NanoLC-MS/MS)技术分离鉴定CC和CAP的差异表达蛋白质,并用生物信息学方法对差异蛋白质在结肠癌变过程中的生物学意义进行分析.结果 共鉴定266个差异蛋白质,其中103个蛋白质在CC上调,163个蛋白质下调,这些差异蛋白质涉及细胞代谢,免疫,运输,发育,细胞黏附,细胞周期,凋亡等过程.结论 上述差异蛋白质的鉴定为结肠癌癌变机制的研究及分子标志物确定提供了有益的科学依据.

Quantitative proteomics analysis of human colon cancer relative to colon adenomatous polyp epithelial cells isolated by laser capture microdissection

Abstract:Objective To investigate the differentially expressed protein profile of colon cancer (CC) relative to colonic adenomatous polyp (CAP).Methods Laser capture microdissection (LCM) was used to obtain colon cancer and adenomatous polyp epithelial cells.Isobaric tags for relative and absolute quantitation (iTRAQ) and nano-liquid chromatography-tandem mass spectrometry (NanoLC-MS/MS) were used to isolate and identify differentially expressed proteins between CC and CAP.Bioinformatics was used to mine the biological significance of differentially expressed proteins in the colon carcinogenesis processes.Results A total of 266 differentially expressed proteins was identified,of which 103 proteins were up-regulated and 163 down-regulated in CC.Those proteins were involved in cell metabolism,immunity,transport,development,cell adhesion,cell cycle,and apoptosis,etc.Conclusions Those differentially expressed protein data might be a useful scientific basis for the clarification of cancer mechanisms and the discovery of molecular markers.
