^32磷——玻璃微球对动物肝脏伤害性的观察

摘    要:目的肝动脉内给予32磷-玻璃微球(32P-GMS)对家猪肝纤维化影响的评估。方法10只健康家猪经皮肝动脉内分别接受459MBq~1070MBq的31P-GMS(5只猪)、31P-GMS(5只猪)及造影剂(2只猪)。于术后第1、2、4、8或16周分批处以安乐死,暂不处死的同期做肝脏穿刺,取肝组织进行超微结构观察。存活猪每周检测血清肝纤维化标志物。结果家猪安乐死时全肝或某肝叶组织所接受的宏观平均吸收剂量范围为48~190Gy。组织学表现肝脏损伤修复的时间需8周以后。血清学表现透明质酸(HA)和人Ⅲ型前胶原(hPCⅢ)术后1、2周有一过性增高,8周后恢复正常。结论家猪肝动脉灌注人类治疗剂量的32P-GMS所至肝损伤为可逆性,观察120天没有发现肝纤维化变。血清HA和hPCⅢ测定是监控给药后肝脏功能的重要量化指标。

Abstract:Objectives To evaluate the influence of percutaneous intrahepatic arterial administrated 32 P-glass microspheres (32 P-GMS) with the dosage equivalent to the therapeutic dose in human beings on the liver of domestic pigs. Methods 32 P-GMS ,459MBq1070MBq was administrated by percutaneous intrahepatic arterial catherization to 5 healthy domestic pigs, and 31 P-GMS or contrast agent were given by the procedure same as above to other 5 or 2 healthy pigs respectively. One pig from each group was surrendered to euthanasia at 1st ,2nd ,4th, 8th or 16th wk. Liver biopsies were performed on the rest pigs at the corresponding time point.The ultrastructures of liver tissue specimens were examined. Blood specimens were taken weekly from the surviving pigs for determining the liver function parameters. Results At the time of euthanasia ,the whole liver or some lobe tissues received a macrocosmic absorbed average dose ranged from 48 Gy 190Gy.The duration required for the injured liver tissues to be repaired hepatic artery/animalLiver fibrosiswas more than 8 wks. The serological manifestations of hyaluronic acid and human procollagen (hPC) had a transient elevation at 1st or 2nd wk postoperation ,then restored to normal at 8th wk. Conclusion After 120 day's observation, the pigs received reasonable dose of 32 P-GMS by intrahepatic arterial administration ,we have not found prominent evidence of liver injury neither in histology nor in liver function.
