再生障碍性贫血中色氨酸和犬尿氨酸的检测方法及意义

摘    要:目的 建立检测再生障碍性贫血(Aplastic Anemia,AA,)血清中必需氨基酸——色氨酸(Trp)及其代谢产物犬尿氨酸(Kyn)水平的方法,探讨其与疾病之间的关系。方法 应用反向高效液相色谱法检测,以荧光检测器检测色氨酸、以紫外检测器检测犬尿氨酸的血清水平 结果 再障患者血清Trp浓度明显低于正常对照,Kyn浓度明显高于正常对照;再障动物模型结果与患者血清结果相同(P<0.05)。结论 在再障发病过程中,色氨酸限速酶IDO的表达水平和活性均较正常明显增高,促进色氨酸向犬尿氨酸转化,色氨酸的不足导致细胞内的“色氨酸饥饿状态”从而抑制T细胞的活化和增殖,是机体免疫系统自我调节的一种重要保护机制。

Determination of Tryptophan and Kynurenine in Aplastic Anemia and its significance
Abstract:objective: To establish the method for the simultaneous determination of tryptophan and kynurenine in serum of aplastic anemia patients, study the relationship between the changes of tryptophan catabolism and the disease. Methods Tryptophan and kynurenine concentration were detected by high-performance liquid chromatography (HPLC) Results: Tryptophan concentration in patients of AA was lower than control significantly(P<0.05),while kynurenine in serum was higher than control.(P<0.05).The results were same with mouse model. Conclution: The deficiency of the essential amino acid tryptophan in AA caused the status of “ tryptophan starve” which could inhibit the proliferation of Th1 cells, then promoted the recovery of the disease. IDO expression level and tryptophan concentration is important for pathogenesis and treatment strategy.
