| 摘 要： | 单克隆抗-HBs在同系BALB/c 小鼠体内所诱生的抗独特型抗体,能模拟HBsAg的作用,该抗独特型抗体既能与抗-HBs发生特异性结合,又能在同系动物中添生出抗HBs的独特型抗体,从而为生产乙肝疫苗开辟了一条新的可能的途径。由于单克隆抗-HBs在同系动物体内只诱生抗独特型抗体,故无同种型和同种异型的干扰,且纯化程序简单,避免了许多干扰因素,有利于研究机体免疫网络中各细胞间的相互作用。

| Abstract: | Anti-idiotype antibodies (Ab2) weresuccessfully induced by monoclonal anti-HBsantibodies (Ab1) in syngenic BALB/c mice.They(Ab2)were identified by RPHA, ELISAand BASA tests. The results showed thatthe Ab2 were able to combine not onlywith monoclonal anti-HBs from mouse butalso with polyclonal anti-HBs antibodiesfrom horse.It was suggested that a commonantigenic determinant existed both in mo-noclonal anti-HBs from mouse and in poly-clonal anti-HBs antiserum from horse. Asmonoclonal antibodies can only induce anti-idiotype antibodies in syngenic animals, theinduction of Ab2 with monoclonal Ab1 insyngenic mice is more convenient than thatwith polyclonal Ab1 in different animals.It is a simple and reliable method for pre-paration of Ab2, and it may be suitablefor the study of regulations of idiotypeand anti-idiotype network within bodies.By using Ab2 thus prepared to immunizesyngenic mice,we obtained anti-anti-idiotype antibodies(Ab3 or Ab1') with the spe-cificity of anti-HBs. It is revealed that theidiotype of prepared Ab2 could mimic theantigenic determinant of HBsAg. It seemslikely that Ab2 might be used as a newvaccine for the protection of HBV infection. |
