PSTAT3与VEGF在胰腺癌中的表达及其临床意义

目的探讨PSTAT3和VEGF蛋白在胰腺癌组织中的表达情况及其与胰腺癌临床病理特征、胰腺癌组织中血管新生的关系.方法用免疫组织化学检测了41例手术切除的胰腺癌组织和10例正常胰腺组织中PSTAT3、VEGF的蛋白表达和微血管密度.结果在41例胰腺癌标本中有29（70.7%）例PSTAT3和31（75.6%）例VEGF蛋白的表达阳性,明显高于正常胰腺组织.PSTAT3和VEGF蛋白的阳性表达呈正相关（r=0.758,P＜0.01）.PSTAT3在临床分期较晚和有淋巴结转移的胰腺癌中呈高表达（P=0.003,0.033）,VEGF的表达与临床分期有关（P=0.025）.41例胰腺癌组织中的微血管密度MVD与PSTAT3、VEGF蛋白的阳性表达有关（P=0.003,0.001）.结论胰腺癌组织中存在PSTAT3和VEGF的过表达,PSTAT3与临床病理分期和淋巴结转移有关,PSTAT3可能通过上调VEGF的表达,促进胰腺癌组织中新生血管的形成,可能在胰腺癌的发生发展中起到重要作用.

Expression of PSTAT3 and VEGF in pancreatic carcinoma and its clinical significance

Objective To investigate the expression of PSTAT3 and VEGF in human pancreatic carcinoma and explore its relation to clinicopathological features and tumor angiogcnesis.Methods The expression of PSTAT3 and VEGF was determined with immunohistochemistry in samples of tumor tissue from 41 patients with pancreatic carcinoma and those of pancreatic tissue from 10 healthy individuals.The MVD was also measured by immunohistochemistry using CD34 antibody.Results The expression rates of PSTAT3 and VEGF in pancreatic carcinoma tissue were 70.79%(29/41)and 75.6%(31/41),respectively.They were significantly higher than those in the normal group.The positive rate of PSTAT3 was closely related to clinical stage and lymph node metastasis(P=0.003, 0.033).There was positive correlation between the positive rate of PSTAT3 and that of VEGF(r= 0.758.P<0.01).VEGF was also associated with clinical stage(P=0.025).In the 41 specimens of pancreatic carcinoma,the increased MVD was related to positive expression of PSTAT3 and VEGF(P =0.003,0.001).Conclusions PSTAT3 and VEGF are over-expressed in human pancreatic carcino- ma.VEGF might be regulated by PSTAT3 to result in tumor angiogenesis.Therefore,PSTAT3 may play an important role in the carcinogenesis and progression of pancreatic carcinoma.
