脊髓星形胶质细胞机械性损伤的体外培养模型

摘    要:目的:建立机械性损伤引起星形胶质细胞反应性增生的体外培养模型,方法:体外分离培养新生Wistar大鼠脊髓的星形胶质细胞,培养至16d进行划伤实验,分为划伤组和对照组(未划伤);每组有7个时间点,分别为划伤后0,2h,6h,12h,24h,48h,72h。用免疫组织化学的方法进行染色,观察划伤组和对照组星形胶质细胞内胶质纤维酸性蛋白(GFAP)的表达;并且进行半定量图像分析。结果:从划伤后2h-72h,同对照组相比,划伤组星型胶质细胞明显增生,细胞内GFAP的表达明显增高。结论:星形胶质细胞反应性增生的体外培养划伤模型能模拟髓组织受到机械损伤后,胶质细胞反应性增生及细胞内GFAP表达升高的过程。

Abstract:Objective:To build the model of astrocytic reaction after mechanical injury in culture. Methods: The astrocytes of spinal cord of new born WISTAR rats were cultured in vitro. After fifteen days, we nicked the bottom of the cell culture dishs by tips. Two groups were divided, one is control group, the other is injured group. There are seven time points in each group, such as 0h,2h, 6h, 12h, 24h,48hrs,72h. Immunocytochemistry was used to analysis the expression of GFAP of astrocytes of the two groups. Result: The expression of GFAP of the astrocytes of injured group is apparently increased. Conclusion: The result demonstrates that the model is successful.
