丹参酮ⅡA对佐剂型关节炎大鼠离体胸主动脉环的舒张作用及对外周血vWF表达的影响

目的：研究丹参酮ⅡA（TanⅡA）对佐剂性关节炎（AA）大鼠离体血管的内皮依赖性舒张作用及外周血管假性血友病因子（vWF）表达的影响。方法：建立AA大鼠模型并随机分组，各组连续给药3周后处死大鼠。采用离体血管环灌流讲述观察各组给予去甲肾上腺素（NA）预收缩后加入乙酰胆碱（Ach）的内皮依赖性舒张作用，同时酶联免疫吸附法（ELISA）检测外周血vWF含量。结果：与空白对照组相比，模型组、MTX组血管环应用Ach后对去甲肾上腺素（NA）引起的收缩反应的舒张率显著降低，TanⅡA组与空白组相比差异无显著性；模型组vWF水平最高，与空白对照组、MTX组、TanⅡA组差异具有显著性，TanⅡA组水平最低。结论：AA大鼠主动脉内皮依赖性舒张作用受损机制可能与内皮功能受损有关，TanⅡA可以修复内皮细胞，改善RA心血管损伤。

The Effect of Tanshinone IIA Influence the Isolated Aorta Rings Rat Thoratic Relaxant Response and the Serum Von Willebrand Factor in Adjuvant Arthritis Mice

Objective The aim of this study is to investigate the effect of Tanshinone ⅡA （Tan ⅡA） to the level of von Willebrand Factor （vWF） in serum and relaxant response depend on endothelium of isolated aortas in adjuvant arthritis （AA） mice. Methods Create animal model of adjuvant arthritis and randomization, after 3 weeks administration kill the mice and get serum, vWF was assayed by ELISA. Determining relaxant response depending on endothelium after adding Ach in organ bath with NA pre-contracted. Results The acetylcholine （Ach） induced endothelium dependent relaxation are decreased in aortas from model group, MTX group. But there is no significant difference between Tan ⅡA group and control group, vWF： Model group have the highest vWF level, has the significant difference comprared with control group. MTX group and Tan ⅡA group, Tan ⅡA group are lower than the other group. Conclusion Tan ⅡA can improve cardiovascular damage of Rheumatoid arthritis.
