BA-ELISA快速检测鼻咽分泌物中合胞病毒抗原的研究

摘    要:本文采用自制的RSV免疫血清以及国产试剂,通过特异性、敏感度和重复性试验,建立了检测RSV抗原的生物素-亲和素-酶联兔疫吸附试验(BA-ELISA)。该法比常规的ELISA法敏感4倍。用此法检测58份肺炎和毛细支气管炎患儿鼻咽分泌物(NPS)中的RSV抗原,结果与组织培养(TC)分离RSV结果比较:其特异性和敏感性分别为91.2%和95.8%,两法诊断符合率为93.1%。结果表明本法不但敏感、特异,而且简便、快速、经济,在收到标本24小时即可得到结果,是一种适应于基层推广的RSV感染诊断方法。

Abstract:A biotin-avidin-enzyme-linked immunosorbent assay for detection of respiratory syncytial virus (RSV) antigens was developed, using antisera made in our institute. Guinea pig and rabbit anti--RSV immunoglobulins were uSed as capture and detector antibodies respectively.The specificity of this method was determined by crossing and blocking tests, revealing the sensitivity being 4 times higher than that ofconventional ELISA.Applying the assay to decteet RSV in 58 NPS specipsens, thespecficity and sensitivity were 91. ZN and 95.8% respectively.The result showed thatthe assay could be easily performed in 24h and was highly sensitiye and specific.
