摘    要:本文叙述了用反相色谱法,采用三元溶剂体系分离测定生物样品中三种腺苷酸.三元溶剂为水、甲醇、乙腈(80∶15∶5)含0.005MpicA.在室温下等度洗脱能使AMP,ADP,ATP标准物25min完全分离.大鼠肝组织提取液中的AMP,ADP,ATP也能与杂质完全分离.再现性好,并优于Harmson和Rao报道的方法.五次测定结果的变异系数为AMp1.9%,ADP4.5%,ATP6.5%.回收率为95%、101%、99%.该方法能对大鼠肝、心肌、骨胳肌等组织样品中腺苷酸同时进行准确的测定.

Abstract:A method for the quantitative determination of adenine nucleotides in biological materials by reversed phase high-pressure liquid chromatography is described.A solution system of water:methanol:acetonitrile (80:15:5) containing 0.005mol picA/L was used to separate AMP,ADP and ATP.The chromatogram may be completed in 23 minutes.AMP,ADP and ATP were quantitated by the peak height/weight ratio of the standard materials.The coefficients of variation of five determinations of AMP,ADP and ATP in rat liver samples were 1.9%,4.46% and 6.5% and their mean recovery rates were 97%,101% and 99%,respectively.This method is simple,rapid,reproducible,economic,and useful in laboratory.
