动脉血栓靶向超声显像的体内实验研究

摘    要:目的探讨白蛋白血栓靶向超声造影剂体内增强动脉血栓超声显像的效果.方法采用FeCl3溶液诱发30只健康新西兰大白兔腹主动脉非梗阻性新鲜血栓形成;经耳缘静脉团注0.04 ml/kg的白蛋白超声造影剂进行超声造影,分为靶向组(20只)和非靶向组(10只);采用视觉观察评价血栓显像增强效果.结果 29只(29/30)兔模型建立成功;造影后视觉观察,靶向组18例达到2级增强效果,1例为1级增强效果,1例死亡;非靶向组仅2例达到1级增强效果,两组比较差异显著(P<0.01).结论白蛋白血栓靶向超声造影剂可显著增强兔腹主动脉内新鲜血栓超声显像效果.

Abstract:Objective To study method of non obstructive thrombosis in artery, and to evaluate the enhancement effect of the thrombus targeted ultrasound contrast agent on acute thrombus in normal rabbit abdominal aorta. Methods Non obstructive thrombus resulted with FeCl 3 in abdominal aorta of 30 rabbits was formatted. Ultrasound images were obtained before and after administration of microbubbles. Healthy rabbits were injected contrast agent at ear by intravenous bolus injection of the albumin targeted and non targeted ultrasound contrast agent in a dose of 0.04 ml/kg, respectively. The images were qualitatively assessed by three independent observers to determine the contrast enhancement effect of acute thrombus as grade 0 (no enhancement), grade 1 (slight enhancement effect)and grade 2 (obvious enhancement effect). Results Non obstructive thrombosis in artery was formatted successfully in 29 cases. After the albumin targeted ultrasound contrast agent was injected, there were 18 cases whose thrombus were enhanced brilliantly as grade 2 while only 1 case as grade 1. The obviously enhanced effect of the acute thrombus lasted more than 10 minutes. While after non targeted ultrasound contrast agent was injected in 10 cases, the clots were generally difficult or impossible to detect ( P <0.01). Conclusion Method of non obstructive thrombus resulted with FeCl 3 in abdominal aorta is convenient. Thrombus targeted albumin microbubbles can improve ultrasound imaging of acute thrombus in rabbit abdominal aorta.
