风湿康治疗大鼠膝骨关节炎的实验研究

摘    要:目的探讨风湿康治疗大鼠膝骨关节炎的作用机制。方法膝关节腔注射4%的木瓜蛋白酶建立大鼠OA模型,1周后48只大鼠随机平均分成正常组、模型组、双醋瑞英组及1.35g/kg、2.7g/kg、5.4g/kg的风湿康治疗组共6组,正常组与模型组给与等体积的生理盐水。4周后处死动物,检测各组大鼠血清中IL-1β和TGF-β1的含量,切取大鼠膝关节,行大体和组织学观察,采用免疫组织化学方法检测基质金属蛋白酶-13(MMP-13)在关节软骨中的表达。结果风湿康治疗组均能降低血清中IL-1β的水平(P〈0.001),风湿康中剂量和高剂量组能显著升高血清中TGF-β1的水平(P〈0.001)。风湿康低剂量组对关节软骨的保护作用并不明显(P〉0.05),风湿康中剂量和高剂量组均可显著抑制MMP-13在膝关节软骨中的表达(P〈0.05),风湿康高剂量组与双醋瑞英组具有相似的疗效(P〉0.05)。结论风湿康可能是通过降低血清中IL-1β、升高TGF-β1的水平、减少MMP-13在膝关节软骨中的表达,从而发挥其对骨关节炎的治疗作用。

Effect of Feng Shi Kang on experimental osteoarthritis in rats

Abstract:Objective To investigate the effect of feng shi kang (FSK) on experimental knee osteoarthritis in rats. Methods Forty-eight rats were equally divided into six groups:normal group, model group, Diacerein group, and FSK groups receiving 1.35,2.7,5.4 g/kg of FSK.Both the normal and model groups were treated with normal saline. Animal model of osteoarthritis was established by intra-articular injection of 4% papain in the knee of the rats, and the rats were then treated with FSK at 1 week after papain injection. The animals were treated with FSK daily for 4 weeks. The levels of serum IL-1β and TGF-β1 were determined by ELISA at 4 weeks after FSK treatment. Macroscopic and histological examinations were performed on the articul arcartilage. Immunohistochemical and morphological analysis were performed to examine the in situ expression and distribution of matrix metalloproteinases-13(MMP-13) in the cartilage. Results FSK was found to reduce the production of IL-1β(P0.001). A moderate to high concentration of FSK could significantly increase the levels of TGF-β1 (P0.001). Low concentration of FSK could not significantly repair cartilage degeneration(P0.05). A moderate to high concentration of FSK could significantly inhibit the expression of MMP-13 in the articular cartilage(P0.05). High concentration of FSK showed a similarly inhibitory effect as Diacerein in the inhibition of MMP-13 expression(P0.05). Conclusions FSK was found to exhibit anti inflammation effect and protect articular cartilage. The effect was correlated with the reduction of IL-1β, an increasing in the level of TGF-β1 in the serum, and the reduced expression of MMP-13 in the articular cartilage.
