端粒酶RNA反义寡核苷酸对人肺癌细胞端粒酶活性和细胞增殖的影响

摘    要:目的 观察脂质体介导的端粒酶RNA反义寡核苷(ASODN)对人肺癌细胞系端粒酶活性的作用及其对癌细胞增殖的影响。方法 合成与人类端粒酶RNA模板区碱基序列互补的ASOND,并用阳离子脂质体转染剂DO-SPER介导,作用于人肺癌细胞系,空白组和正义寡脱氧核苷酸(SODN)作为对照。用以PCR为基础的端粒重复序列扩增法(TRAP)结合ELISA方法检测肺癌细胞的端粒酶活性。台盼蓝拒染法细胞计数、噻唑蓝(MTT)试验观察癌细胞增殖情况。结果 脂质体介导的抗端粒酶ASODN对肺癌细胞的端粒酶活性具有抑制作用;细胞计数和MTT试验显示肺癌细胞增殖速度明显减慢。结论 针对端粒酶模板区的ASODN可显抑制端粒酶活性和肺癌细胞的增殖,可运用于抗肿瘤实验性治疗和相关研究。

Abstract:Objective To study the effect of specific antisenseoligonucleotides of telomerase on the telomerase activity and lung carcinoma cell proliferation Methods Antisense and sense oligonucleotides(ASODN and SODN) was synthesized according to the sequence of the template region of RNA component of human telomerase Lung Carcinoma Cells were treated with ASODN and SODN mediated by DOSPER Telomerase activity was detected by PCR based telomeric repeat amplification protocol(TRAP) combined with ELISA Number of cells were counted by trypanblue staining Inhibiting rate deteminted by MTT assay Results The ASON reduced the telomerase activety (P<0 001) and the number of cells (P<0 01) and elevated the inhibiting rate (P<0 001) as compared with SODN and the empty controls Conclusion ASON targeted the RNA template inhibited the telomerase activity and the lung cell proliferation and therefore potentially might be significant in tumour therapy
