透明质酸凝胶在成肌细胞体外培养中的应用

摘    要:目的探讨透明质酸凝胶与成肌细胞的生物相容性,为成肌细胞组织工程研究提供新型载体。方法将体外培养的成肌细胞接种于含透明质酸凝胶的培养液中常规培养,通过倒置显微镜和扫描电镜观察细胞形态,描绘细胞生长曲线,并进行免疫细胞组化鉴定。结果透明质酸凝胶组与对照组成肌细胞体外培养生长曲线相似,均成desmin阳性,透明质酸凝胶组亲水性和吸附力优于对照组。扫描电镜下透明质酸呈三维框架结构,有利于成肌细胞附着。结论透明质酸凝胶是成肌细胞良好的载体材料。

Abstract:Objective To explore the biocompatibility between hyaluronic acid gelatum and myoblast to provide novel carrier for myoblast tissue engineering research. Methods Myoblasts were seeded in the culture medium containing hyaluronic acid gelatum and cultured as routine. The morphological properties of these cells were observed with inverted microscope and scanning electron microscope(SEM). The proliferative curve of the cells was described and the specificity was identified by immunocytochemistry with myoblasts specific marker of anti-desmin. Results The proliferative curve of myoblasts was similar between hyaluronic acid gelatum group and control group with all myoblasts desmin positive. While the hydrophilicity and absorption capacity of myoblasts cultured in hyaluronic acid gelatum were much better than those in control group. The ultrastructure of myoblasts in hyaluronic acid gelatum showed three dimensional frameworks under SEM which was beneficial for myoblasts adhesion. Conclusion Hyaluronic acid gelatum is an excellent carrier material for myoblasts culture.
