亚硒酸钠与白藜芦醇联用对肺癌细胞抗氧化和细胞周期蛋白的影响

摘    要:目的:研究亚硒酸钠与白藜芦醇联用对肺癌细胞抗氧化和细胞周期蛋白的影响,从而探讨其对肺癌细胞存活和凋亡的作用。方法:采用细胞培养技术培养肺癌A549细胞,分别将细胞分为对照组、亚硒酸钠组、白藜芦醇组、亚硒酸钠和白藜芦醇联用组,采用MTT法检测细胞存活率,流式细胞术检测凋亡率和ROS水平,用酶标仪检测谷胱甘肽(GSH)和丙二醛(MDA)表达水平,Western blot法检测细胞周期蛋白(P-Rb)表达水平。结果:与单独处理组相比,亚硒酸钠与白藜芦醇联用可降低肺癌A549细胞的存活率,促进其凋亡,且与对照组比较差异有统计学意义(P < 0.05);同时,与单独处理组相比,联用组能显著降低GSH表达水平(P < 0.05)和显著增加MDA (P < 0.05)和ROS表达水平(P < 0.05),联用组可以显著降低周期蛋白(P-Rb蛋白)的表达水平(P < 0.05)。结论:亚硒酸钠和白藜芦醇的联用对肺癌细胞有抑制增殖、促进凋亡的作用,其机制可能与增加氧化损伤及降低P-Rb蛋白表达有关。

Abstract:OBJECTIVE: To investigate the effect of sodium selenite in combination with resveratrol on anti-oxidant and cyclin in lung cancer A549 cells,and on cell survival and apoptosis. METHODS: A human lung adenocarcinoma cell line,A549,was established and divided into several groups:control,sodium selenite,resveratrol,and combined sodium selenite and resveratrol groups. Methyl thiazolyl tetrazolium (MTT) method was performed to assess proliferation of A549 cells. Cell apoptosis rate and ROS were detected by flow cytometry. Glutathione (GSH) and malondialdehyde (MDA) were detected by Microplate Reader. The protein expression of P-Rb was detected using Western blotting. RESULTS: Compared with the control group,treatment with the combination of sodium selenite and resveratrol significantly reduced cell survival and increased apoptosis (P < 0.01). Within these cells,the content of GSH was significantly reduced (P < 0.05), and MDA and ROS were significantly increased (P < 0.05). The protein expression of P-Rb was down-regulated (P < 0.05). CONCLUSION: Treatment with the combination of sodium selenite and resveratrol inhibited proliferation,induced apoptosis which were associated with increased oxidative damage and down-regulating protein expression of P-Rb.
