摘    要:目的优化叠鞘石斛多糖超声提取工艺。方法以3年生叠鞘石斛为材料,采用超声辅助法对新鲜叠鞘石斛茎段中的多糖成分进行提取,苯酚-硫酸法检测多糖含量,在单因素试验基础上,以超声功率、超声温度、提取时间为影响因素,得率为响应值,采用Box-Behnken响应面法,优选叠鞘石斛多糖最佳超声提取工艺技术参数。结果与结论试验结果表明,叠鞘石斛多糖最佳提取工艺条件为:超声功率320W、温度80℃、时间20min,在该条件下,理论多糖提取率为27.53%,经3次重复验证试验得到的多糖提取率平均值为27.05%,与理论值的误差仅为1.74%。

Abstract:Objective To optimize the ultrasonic extraction technology of polysaccharides from Dendrobium denneanum. Methods The ultrasonic extraction method of polysaccharides from Dendrobium denneanum was optimized by using three-year-old fresh branches as materials. The Phenol Sulphuric acid method was employed to detect the content of polysaccharides. On the base of single factor tests, the Box-Behnken Response Surface Analysis was adopted to optimize polysaccharides extraction technology technical parameter. Extraction rate was regard as response value and the dosage. Ultrasonic power, extraction temperature, and extraction time were done as factors. Results and Conclusion The optimum conditions for ultrasonic extraction of Dendrobium denneanum were as follows: ultrasonic power 320W, temperature 80℃ and time 20min. Under these conditions, the theoretical extraction rate of polysaccharide was 27.53%, the average extraction rate of the three repeated experiments was 27.05%, and the theoretical value of the error was only
