摘    要:目的:探讨p15基因在食管癌中的表达以及与食管癌临床病理及预后的关系。方法:应用酚:氯仿法提取46例食管癌患者的肿瘤及癌旁正常组织中高分子量的基因组DNA,并对p15基因外显子1和2进行PCR扩增,以1.5%琼脂糖凝胶电泳及溴化乙烷染色,分析PCR产物中p15基因的纯合性缺失。结果:正常食管粘膜组织中未见p15基因的纯合性缺失。食管癌肿瘤组织中,p15基因的纯合性缺失率为63.04%(29/46),且与食管癌的临床病理分级及DNA含量有关。结论:p15基因的纯合性缺失在食管癌发生、发展中起重要作用,可作为肿瘤标志物判断食管癌患者的预后。

Abstract:Objective: To investigate the expression of p15 gene in esophageal cancer and its relationship with clinical pathologic parameters and prognosis. Methods: High molecular weight genomic DNA isolation was performed by using standard phenol and chloroform extraction. PCR methods were used with primers which were specific for exon l and 2 of the p15 gene. PCR products were stained with ethidium bromide and run on 1.5% agrose gel for p15 gene homozygous deletion. Results: There were no p15 gene homozygous deletions in normal esophageal mucosal. p15 gene homozygous deletion rate in esophageal cancer was 63.04%(29/46) and it associated with low grade esophageal cancer and DNA contents. Conclusion: p15 gene homozygous deletions played an important role in the development and progression of esophageal cancer and could be used as a tumor marker to assess the prognosis of esophageal cancer.
