超支化聚醚酯-二氢卟酚e6纳米光敏剂的构建与表征

摘    要:目的:研究超支化聚醚酯-二氢卟酚e6纳米光敏剂[hyperbranched poly (ether-ester)-chlorin e6 nanophotosensitizer,HPEE-ce6]的构建过程与表征,及其对人舌鳞癌CAL-27细胞(Tongue cancer CAL-27 cells)体外光动力疗法(Photodynamic therapy,PDT)的杀伤作用.方法:通过一步台成法构建HPEE-ce6纳米光敏剂,用紫外可见吸收光谱和透射电镜观察其表征,通过透射电镜观察CAL-27细胞的形态学改变.结果:HPEE和ce6共价结合,生成直径为50 nm左右的纳米粒子,其治疗区吸收峰比ce6红移了12 nm;经PDT处理后,形态学研究表明,CAL-27细胞发生凋亡或特征性肿胀样坏死.结论:HPEE-ce6可以通过一步法(HPEE和ce6共价交联)合成,并对人舌癌CAL-27细胞具有良好的光动力杀伤作用.
