摘    要:目的观测年龄对人骨髓间充质干细胞(human mesenchymal stem cells,hMSCs)体外增殖、成骨分化的影响.方法使用密度梯度离心法分离不同年龄人骨髓MSCs进行培养,保留贴壁细胞传代,观察细胞生长情况,检测其增殖活性、细胞周期、碱性磷酸酶活性.结果低龄hMSCs较高龄hMSCs体外生长快、增殖期比例大、MTT及碱性磷酸酶活性高.结论hMSCs的增殖和成骨分化的能力和活性随着年龄的增加而降低,建立其参数可满足组织工程种子细胞在临床应用中不同患者的要求.

Abstract:Objective To determine whether the donor age influences proliferation and differentiation capability of human mesenchymal stem cells (hMSCs). Methods HMSCs cultures were obtained by using density centrifuge method from 39 donors aged from 7 to 79 years old. The adhesive cells were preserved to passage culture, then the proliferation and osteogenic differentiation capability were investigated. Results The total culture time increased with the age of donors. Cell cultures from younger donors showed higher activity of MTT and expression of alkaline phosphatease (ALP) than older donors. Conclusion The proliferation and osteogenic differentiation capability of hMSCs decreases with the age of donors. Our study has highlighted the significant effect of both donor age and in vitro expansion on hMSCs quality. These findings have significant implications for the proposed clinical application of hMSCs.
