茵陈蒿汤对肝内胆汁淤积湿热证大鼠肝组织NTCP表达的影响

摘    要:目的 研究茵陈蒿汤对肝内胆汁淤积湿热证大鼠肝组织中钠-牛璜胆酸盐共转运多肽( Na+/taurocholate Cotransporting Polypeptide,NTCP)表达的影响,探讨茵陈蒿汤的作用机制.方法 选用雄性SD大鼠45只,随机分为3组:正常对照组、模型对照组和茵陈蒿汤组.观察大鼠的体质量、肝功能、肝组织病理变化和肝脏NTCP的表达.结果 治疗后茵陈蒿汤组大鼠的体质量为( 196.33±3.42)g,高于模型对照组(182.80±5.89)g(P<0.05)].茵陈蒿汤组大鼠血清TB、DB、ALT、AST、ALP水平分别为(13.90±2.88 )μmol·L-1、( 7.47 ±2.70) μmol·L-1、(75.33±9.42)U ·L-1、(216.64土31.20)U·L-1、(226.40±32.86) U· L-1,均低于模型对照组(34.72±3.65 )μmol·L-1、(19.33±3.27)μmol·L-1、(123.73±18.51)U·L-1、(353.78±63.08)U·L-1、(355.49±63.08)U·-1(P<0.05).苏木精-伊红染色法显示茵陈蒿汤组大鼠肝细胞的坏死、炎症、水肿程度与模型对照组组比较均有减轻.免疫组化染色示茵陈蒿汤组大鼠肝细胞NTCP阳性表达高于模型对照组.荧光定量RT-PCR检测显示茵陈蒿汤组大鼠肝细胞NCTP mRNA表达(2-△△CT值为2.38±0.76)高于模型对照组(2-△△CT值为1.48±0.38)(P<0.05).结论 茵陈蒿汤能增加肝内胆汁淤积湿热证大鼠的体质量,有效改善胆汁淤积、肝功能及减轻肝脏病理损害.茵陈蒿汤能提高肝内胆汁淤积湿热证大鼠肝细胞NTCP的表达.

Abstract:Objective To investigate the therapeutic mechanism of Yinchenhao Decoction(YD) by observing the effect of YD on hepatic Na+/taurocholate cotransporting polypeptide(NTCP) expression levels in rats with intrahepatic cholestasis and damp-heat syndrome.Methods Forty-five male SD rats were randomly assigned into 3 groups:normal,model control and YD group.The body weight,liver function,liver pathological changes and hepatic NTCP expression in rats were detected.Results The rat body weight in YD group was(196.33±3.42)g,greater than(182.80±5.89)g in model control group after treatment(P<0.05).The serum levels of TB,DB,ALT,AST,ALP in YD group(13.90±2.88 μmol·L-1,7.47±2.70 μmol·L-1,75.33± 9.42 U·L-1,216.64±31.20 U·L-1,226.40±32.86 U·L-1,respectively) were lower than those in the model control group(34.72±3.65 μmol·L-1,19.33±3.27 μmol·L-1,123.73±18.51 U·L-1,353.78±63.08 U·L-1,355.49±63.08 U·L-1,respectively)(P<0.05).The results of hematoxylin-eosin staining showed that the degree of necrosis,inflammation and edema of rat hepatocytes in YD group was milder than those in the model control group.More NTCP positive hepatocytes were found in rats of the YD group than those of model control group after immunohistochemical staining.The results of fluorescent quantitative RT-PCR analysis showed that NTCP mRNA expression level in rat hepatocytes of YD group(the value of 2-△△CT being 2.38±0.76)was higher than that of model control group(the value of 2-△△CT was 1.48± 0.38),P<0.05.Conclusion YD can increase the body weight,improve cholestasis and liver function and relieve hepatic pathological damage of rats with intrahepatic cholestasis and damp-heat syndrome,and YD also enhances NTCP expression level in the hepatocytest.
