摘    要:目的 探讨CD154反义RNA诱导Jurkat细胞凋亡的作用。方法 应用RT-PCR扩增人CD154膜外段基因,以及T-A克隆技术和亚克隆技术构建CD154反义RNA的真核表达载体,并将其转染入Jurkat细胞中。应用电镜及流式细胞仪(FCM)检测观察Jurkat细胞的凋亡指标。结果 电镜观察CD154反义RNA转染的Jurkat细胞内可见凋亡小体,并且FCM检测显示一明显的凋亡峰。结论 CD154反义RNA可诱导Jurkat细胞凋亡。

Abstract:Objective To investigate the effects of CD-{154} antisense RNA on Jurkat cell. {Methods} CD-{154} gene extracelluar domain sequence was amplified by RT|PCR. Then, the PCR product was cloned and subcloned with T|A cloning and subcloning techniques. After the vector was transfected into Jurkat cells, apoptotic data were obtained with flow cytometry(FCM) and electron microscope techniques. {Results} Apoptotic bodies were found in the transfeced Jurkat cells under electron microscope; and FCM analysis showed a significant apoptotic peak. Conclusion CD-{154} antisense RNA can induce apoptosis of Jurkat cells.;
