甲氧滴滴涕对小鼠着床期子宫钙结合蛋白D9k基因表达的影响

摘    要:目的:研究甲氧滴滴涕(MXC)对小鼠着床期子宫钙结合蛋白D9k(Calbindin-D9k)基因表达的影响。方法:将真孕及假孕小鼠于孕1-3d(发现阴栓为孕0日)每日给予不同剂量的MXC(0,100,250,500mg/kg)灌胃,对照组(MXC 0mg/kg)只给芝麻油溶剂。采用RT-PCR检测各组孕鼠子宫Calbindin-D9k基因mRNA的表达;免疫组化染色和图像分析技术检测Calbindin-D9k蛋白表达。结果:①孕鼠经MXC染毒3d后,250,500mg/kg MXC组孕鼠子宫Calbindin-D9k mRNA的表达量与对照组相比有显著降低(P〈0.05),然而,100mg/kg MXC组Calbindin-D9k mRNA的表达与对照组相比虽有所减低,但无统计学意义(P〉0.05);②Calbindin-D9k蛋白在孕5日主要表达在子宫内膜上皮细胞和腺上皮细胞。与mRNA的表达水平一致,250,500mg/kgMXC组孕鼠子宫Calbindin-D9k蛋白的表达量与对照组相比显著降低(P〈0.05),而100mg/kg MXC组Calbindin-D9k蛋白的表达与对照组相比无明显差异(P〉0.05)。结论:MXC可以降低小鼠着床期子宫Calbindin-D9k基因的表达,这可能是其干扰胚胎着床的一个毒性机制。
